PurposePathogenesis of rotavirus infection in mice and its mechanism of diarrhea has not been clearly confirmed. The successfull establishment of suckling mice infected with rotavirus diarrhea model, based on it,study on the effect of Qiweibaizhu Powder for regulating the immune function of mice infected with rotavirus, effects on intestinal mucosa biological barrier, mechanical barrier and immune barrier function, clearance rate of rotavirus and improve the clinical symptoms of neonatal mice. At the same time, study on the effect of Qiweibaizhu Powder of rotavirus SA-11 strain in MA104 cells replication.Material and MethodsRotavirus cultured and passaged in MA104 cells, increase the virulence of the virus, virus titers were detected using fluorescent focus assay. The 5 day old Balb/c mice were inoculated titer of 4.5 × 107FFU/mL rotavirus 50 μ L, establish the model of diarrhea. A normal control group for comparison, after infection divided into model group, Qiweibaizhu Powder group and ribavirin group. Oral gavage for 5 consecutive days, determinate the content of IFN-γ, sIgA in serum and sIgA in intestine mucus, observe the diarrhea and weight changes of neonatal mice, determinate RV antigen detection in fecal with ELISA, judge the virus clearance. Paraffin sections were observed with HE staining and pathological changes of small intestinal villi.Using the experimental method of MTT and CPE in the evaluation of Qiweibaizhu Powder on rotavirus SA-11 replication function in MA104 cells.ResultsNeonatal mice infected with rotavirus, their diarrhea rates and diarrhea days decreased, the detection of RV antigen in feces proved positive, which proved neonatal mice model of rotavirus infection was successfully established.Test of focal fluorescence can be relatively accurate for determination of virus titer. Qiweibaizhu Powder can increase the content of neonatal mice infected with rotavirus serum IFN-γ, sIgA and SIgA in intestinal mucus, enhance the immune function of mice, reduce pathological changes of intestinal villi, restore the barrier function.Effectively alleviate mice diarrhea, dehydration symptoms of rotavirus infection, accelerate viral clearance from the small intestine, reduce the diarrhea rate, reduce the diarrhea days.In the case of low concentrations, Qiweibaizhu Powder which has no cytotoxicity may promote cell growth, inhibitate the replication of rotavirus SA-11 strain on MA104 cells.ConclusionsThis study successfully established a mouse model of rotavirus infection, and on the basis of this experiment, the evaluation of curative effect, Qiweibaizhu Powder on infantile diarrhea proved, its role according to the intestinal mucosal barrier function to protect and restore given to confirm. Qiweibaizhu Powder in vivo and in vitro experiments proved its effective role in anti rotavirus infection.
