Effect Of Qiwei Baizhu Powder On The Related Elements Expression On TLR3 Signal Transduction Pathway Of HRV Infetion Suckling Mice

Objective: To investigate the effect of Qiwei Baizhu powder on the relatedsignaling molecules and inflammatory factors expressionon TLR3 Signal Transduction Pathway in IEL and macrophage of HRV infetion Suckling Mice.Methods: 1. 5-day-old suckling mice were lavaged with 10-7HRV suspension for replicating HRV infection diarrhea model.After the modeling,experimental animal were randomly divided into 8 groups by cote:model group(N G),Qiwei Baizhu powder group(B G),the ribavirin group(ZG), inhibitor group(YG), Qiwei Baizhu powder and inhibitor group(BY G), the ribavirin and inhibitor group(ZY G),Agonist group(JG),and normal control group(NG). Model suckling mice in different group wereorally administrated by 50μLcorresponding drug per for one time per day, which N G and MG were lavaged the volume of sterile saline. After administration of 2、 8、16、24、32、48hours,the TLR3、My D88 cotent in serum of suckling mice were detected with ELISA and the TLR3、My D88 m RNA expression in CD4 + and CD8 + T cells in IELof suckling mice ere detected with RT-PCR. 2. In the same way,the content of IL-6、IL-1β、IL-10 、 IFN-α、 IFN-γ、IRF3 and IRF7 in serum were detected with ELISA and the expression of IL-6、IL-17、IL-10 、 IFN-α、 IFN-γ、IRF3 and IRF7 in CD4 + and CD8 + T cells in IEL were detected with RT-PCR. 3. The DNA3.1 PC plasmid was transfected into macrophage cell line(RAW264.7) by cell transfection technique. The growth trend of the transfected cells and phagocytic function of macrophages was respetively detected by MTT colorimetric method and neutral red phagocytosis assay. After Identification of transfected cells in accordance with experimental requirements,HRV infection transfected cells were treated with Qiwei Baizhu powder,and the IL-6、IL-1β、TNF-α 、IP-10、IFN-β m RNA expression were detected with RT-PCR and real-time PCR.Results: 1. Qiwei Baizhu powder lowered the TLR3、My D88 content in serum of HRV infection suckling mice and down-regulated TLR3、My D88 m RNA expression. 2. Qiweibaizhu powder increased IL-10、IFN-α、 IFN-γ、IRF3、 IRF7 in serum,and reduced IL-6、IL-1βin serum of HRV infection suckling mice,up-regulated the expressionlevelof IL-10、IFN-α、 IFN-γ、IRF3、 IRF7 in CD4 +、CD8 +T in IEL and down-regulated IL-6、IL-17 m RNA expression in CD4 +、CD8 +T in IEL of HRV infection suckling mice. 3. After intervene treatment with Qiweibaizhu powder,the expression of IL-6 、 IL-1β 、 TNF-α 、 IP-10 m RNA expression dropped and IFN-βm RNA expression increased in HRV infection RAW264.7 cell.Conclusion: IL-10、IFN-α、IFN-γ、IRF3 、IRF7 expression ofshowed a low level after infection HRV,while TLR3、My D88、IL-6、IL-17 expression showed high. After the intervene of Qiweibaizhu powder,IL-10、IFN-α、IFN-γ、IRF3 、IRF7 expression up-regulated,while TLR3、My D88、IL-6、IL-17 expression down-regulated.It is suggested that Qiweibaizhu powder induced interferon expression through the activation of IFN regulatory factor(IRF3、7),promoted antiviral infection,and thereby alleviated inflammation infection of intestinal mucosal.
