ObjectiveTo oberve the changes of intestinal metagenome in rotavirus enteritis in animal and clinical experiments, for exploring the role of gut flora in Qiweibaizhu power treating rotavirus enteritis.MethodsThere are two stages in the experiment. In the animal experiment,5days of age SPF healthy kunming neonatal mice were randomly divided into3groups, the Qiweibaizhu Power group, the model control group and the healthy group. The mice of the Qiweibaizhu Power group and the control group were infected with rotavirus solution by gavage, then the rotavirus enteritis models were successfully established. After that, the mice in Qiweibaizhu Power group were given Qiweibaizhu Power elixation, the mice in the other groups given saline solution, all by gavage. After3days, the contents of colons of all survival mice were collected to detect metagenomes, and the data obtained were compared among3groups. In the clinical trial, the intestinal metagenomes of a12-month-old infant infected with rotavirus were compared before and after the therapy of Qiweibaizhu Power.ResultsAccording to the results of animal and clinical experiments, the internal structures of gut flora between mice and infant infected with rotavirus is significant different. For example, the dominant bacteria in gut of mice are Proteobacteria, while the dominant bacteria in gut of mice are Actinobacteria. The internal structures of gut flora are significantly different among three groups of mice, and between the before and after the therapy of the infant. As to the chosen key bacteria, the quantity of intestinal bifidobacterium of the Qiweibaizhu Power group is more than that of the model control group, while the quantity of intestinal lactobacillus of the Qiweibaizhu Power group is less than that of the model control group. On the other hand, after therapy, the quantity of intestinal bifidobacterium decreased, while the quantity of intestinal lactobacillus increased.ConelusionQiweibaizhu Power changed the internal structure of gut flora in neonatal mice and infant infected with rotavirus. However, specific bacteria may not be the targets. With the huge difference between mice and infants, functional genomics analysis may be more valuable.
