Study On The Influence And Clinical Effect Of Buzhongyiqi Particles On Serum IL-3 Of Rats With Spleen Deficiency Allergic Rhinitis Based On "Spleen-stomach Theory Of E.N.T.Department"

1 ObjectiveTo observe if Buzhongyiqi particles can be the more effective and safer drugs treating AR on clinical and in experiments and explore and conclu de the theoretical basis of Buqiyizhong particles treating spleen-deficiency AR. 2 Methods.2.1 Clinical ResearchAll cases were from outpatient service of E. N. T. Department, First Affiliated Hospital of Anhui Medical University from March 2015 to October 2015. Sixty cases of spleen deficiency AR patients that conform to the inclusion standards were included in this study and they were randomly divided into Buzhongyiqi particles group(treatment group) and ebastine group(control group), with 30 cases in each group. Two weeks were taken as one disease course. Changes in nasal symptoms, signs and systemic symptoms of two groups during the treatment process were observed and recorded, separately at the time points before the treatment, one week after treatment,two weeks after treatment and one week follow-up visit. To observe if there were any adverse reactions and conduct statistical analysis on the data.2.2 Experimental StudyForty-eight healthy SD rats were selected and after 2~3 days' adaptive raising,they were divided into the normal control group(12 rats), AR model group(12 rats),spleen deficiency syndrome group(24 rats); after the spleen deficiency syndrome group was successfully evaluated and modeled, it was divided into spleen deficiency AR model group and spleen deficiency AR Chinese medicine group, with 12 rats in each group. The double-antibody sandwich ELISA was adopted to draw the standard curves, with the standard concentration as the abscissa and the OD value as the ordinate. According to OD value of the serum sample, its concentration can be checked on the standard curve and serum IL-3 concentration of rats from different groups can be detected. Then SPSS17.0 software package was used to compare and analyze the obtained data.3 Results3.1 Clinical Observed Results3.1.1 One week after treatment, the effective rate of the treatment group was 9.3%while the effective rate of the control group was 21.2%.3.1.2 Two weeks after treatment, the effective rate of the treatment group was 23.6%while the effective rate of the control group was 29.8%.3.1.3 During one week follow-up visits, the effective rate(35.5%) of the treatment group was better than that of the control group(16.9%).3.1.4 One week after treatment, by being tested by Independent-Samples Tests, running nose, turbinate swelling, reduced appetite and anorexia, fatigue and disinclination to talk, loose stool scores P<0.05 and it had statistical significance, which indicated that one week after treatment, the curative effect of the control group was better than that of the treatment group in improving running nose, turbinate swelling, reduced appetite and anorexia, fatigue and disinclination to talk, loose stool.3.1.5 Two weeks after treatment, by being tested by Independent-Samples Tests,sneezing and reduced appetite and anorexia scores P < 0.05 and it had statistical significance, which indicated that after two weeks' treatment, the curative effect of the control group was better than that of the treatment group in improving sneezing and reduced appetite and anorexia.3.1.6 One week after follow-up visits, by being tested by Independent-Samples Tests,sneezing, rhinocnesmus and abdominal distension scores P<0.05 and the difference had statistical significance, which indicated that one week after follow-up visits, the curative effect of the control group was better than that of the treatment group in improving sneezing, rhinocnesmus and abdominal distension.3.2 Experimental Research ResultsSPSS17.0 software was adopted in this study and the following conclusions were obtained by using Independent-Samples Tests method.3.2.1 Comparing the normal control group and AR group, P= 0.028, P< 0.05, and there was statistical significance. Since 30.984±22.836, IL-3 concentration( SX ±) of AR group, was higher than 13.224±12.560, that of the normal control group, it indicated that IL-3 concentration in the serum of rats from AR group was abnormal and the expression was excessive;3.2.2 Comparing the normal control group and spleen deficiency AR group, P=0.003,P < 0.05, and there was statistical significance. Since 44.803±30.662, IL-3concentration( SX ±) of spleen deficiency AR group, was higher than 13.224±12.560,that of the normal control group, it indicated that IL-3 concentration in the serum of rats from spleen deficiency AR group was abnormal and the expression was excessive;3.2.3 Comparing the normal control group and spleen deficiency AR Chinese medicine group, P=0.040,P<0.05, and there was statistical significance. Since 30.075±23.526,IL-3 concentration( SX ±) of spleen deficiency AR Chinese medicine group, was higher than 13.224±12.560, that of the normal control group, it indicated that IL-3concentration in the serum of rats from spleen deficiency AR Chinese medicine group was abnormal and the expression was excessive;3.2.4 By analyzing the values of concentration((?)±S), it can be seen that the serum IL-3 concentration content of the normal group was evidently lower than that of the other three groups. The concentration of the spleen deficiency AR was highest; the spleen deficiency AR Chinese medicine group was obtained by establishing models after the spleen deficiency AR group taking Buzhongyiqi particles while the concentration of the spleen deficiency AR Chinese medicine group was lower than that of AR group and the spleen deficiency AR group. It can be considered that the concentration of the spleen deficiency AR group decreased evidently after taking Buzhongyiqi particles, close to and lower than the concentration of AR group and recovering to the normal group.4 Conclusion4.1 Clinical research conclusion4.4.1 One week after treatment, the curative effects of the control group in improving the runny nose, turbinate swelling, reduced appetite and anorexia, fatigue and disinclination to talk, loose stool were better than that of the treatment group.4.1.2 Two weeks after treatment, the curative effects of the control group in improving sneezing and fatigue and disinclination to talk were better than that of the treatment group.4.1.3 One week after treatment, the curative effects of the treatment group in improving sneezing, rhinocnesmus and abdominal distension were better than that of the control group.4.1.4 Two weeks after follow-up visits, the long-term curative results of the control group were not ideal; the disease condition of patients from this group was repeated and the symptoms were easy to recur; while the disease recurrence rate of the treatment group was lower, with good long-term effect. The curative effects of the treatment group are continuous, stable and long-term, so it is more worth promoting on clinical.4.2 Conclusion of Experimental Study4.2.1 After successful modeling, the expression of rat serum IL-3 was different.4.2.2 Obvious increase of IL-3 expression was detected in the serum of rats from the AR model group and IL-3 expression in the spleen deficiency group had a further increase compared with AR group.4.2.3 The expression of IL-3 in the spleen deficiency AR Chinese medicine group was relatively high, but compared with the spleen deficiency AR group, it had a evident decreasing trend, also lower than AR group.4.2.4 The application of Buzhongyiqi particles can regulate IL-3 expression of spleen deficiency AR rats, so as to achieve the goal of treating spleen deficiency AR and provide experimental evidence for treating spleen deficiency AR by Buzhongyiqi particles.
