Serum Differential Proteomics Research For Precancerosis Of Chronic Atrophic Gastritis With 3 TCM Syndrome

ObjectiveTo analyze the differential expression of serum proteome in precancerosis of chronic atrophic gastritis(CAG)patients with 3 TCM syndrome,select the specific protein markers of them and find the objective indicators of TCM syndrome.MethodsPrecancerosis of CAG patients were typed by syndrome differentiation and brought into 10 cases weakness of the spleen and stomach syndrome(group X),10 cases syndrome of incoordination between liver and stomach(group G),10 cases spleen-stomach damp-heat syndrome(group S)and choose 10 cases healthy people(group H).All selected person serum were separated proteins by two-dimensional electrophoresis(2-DE)to,stained by silver stain method compatible with mass spectrometry,matched and analysed the images through PDQuest8.0 2-D Analysis Software to find the specific proteins,authenticated the proteins by Reverse phase liquid chromatograph-mass spectrometer(RPLCMS)and analysed them by Uniprot and David database.Result1.The authenticated result of specific proteins of precancerosis of CAG with 3 TCM syndrom(1)19 differential proteins(P<0.05)were found by compared serum proteins in precancerosis of CAG patients with health,among them 10 proteins were authenticated.8 of them were increased expressed,while 2 kinds were increased or decreased within 3 syndrom.(2)74 differential proteins(P<0.05)were found by compared serum proteins in precancerosis of CAG patients of X with health,among them 14 proteins were authenticated.13 of them expression were increased,while 1 kind was decreased.(3)60 differential proteins(P<0.05)were found by compared serum proteins in precancerosis of CAG patients of G with health,among them 9 proteins were authenticated.5 of them expression were increased,while 3 kinds were decreased and haptoglobin was not sure.(4)44 differential proteins(P<0.05)were found by compared serum proteins in precancerosis of CAG patients of X with health,among them 7 proteins were authenticated.3 of them expression were increased,while 1 kind was decreased and 3 kinds were not sure.(5)45 differential proteins(P<0.05)were found by compared serum proteins in precancerosis of CAG patients of X with G,among them 5 proteins were authenticated.4 of them expression were increased,while 1 kind was decreased.(6)41 differential proteins(P<0.05)were found by compared serum proteins in precancerosis of CAG patients of X with S,among them 7 proteins were authenticated.5 of them expression were increased,while 2 kinds were decreased.(7)32 differential proteins(P<0.05)were found by compared serum proteins in precancerosis of CAG patients of G with S,among them 6 proteins were authenticated.4 of them expression were increased,while 2 kinds were decreased.2.The preliminary screening of specific proteins of precancerosis of CAG with 3 TCM syndrom(1)The function of specific proteins between precancerosis of CAG and health were inflammatory response,immune reaction,the migrating,regeneration and apoptosis regulation of tumor cells and so on.β2 glycoprotein(β2GP)and clusterin might be the markers of precancerosis of CAG among them,for their functions were in close relationship with the generation,progression and prognosis of tumor.(2)Inter-alpha-trypsin inhibitor heavy chain H4(ITIH4)was obviously decreased in precancerosis of CAG of X compared with other groups.It might be the marker of precancerosis of CAG of X.(3)Haptoglobin was obviously decreased in precancerosis of CAG of G compared with group X and S.It might be the marker of precancerosis of CAG of G differ from group X and S.(4)Both Ig mu chain C region and protein AMBP were decreased in precancerosis of CAG of S compared with group X and G,while ITIH4 was increased.They might be the markers of precancerosis of CAG of S differ from group X and G.Conclusion1.The specific proteins between CAG with precancerous lesions and health might be associated with the pathogenesis of CAG with precancerous lesions.It might be important to verify them for diagnosing precancerosis of CAG.2.The specific proteins between the syndrome X,G and S of CAG with precancerous lesions may be internal material basis of external manifestation of syndrome.But they were remained to be further verified.
