Effect Of Compound "Lingzhi Jianshen Decoction" On Expressions Of AGT And IGF-1 In Rats With Diabetic Nephropathy

Objective to investigate the clinical efficacy and mechanism of compound "Lingzhi Jianshen Decoction" on expressions of AGT and IGF-1 in diabetic nephropathy rats.Methods The rats with DN were established by injection of small dose streptozotocin(STZ) and divided into the model group(STZ), the treatment group(STZ+ LZJST)and the blank group. The treatment group was teated with compound "Lingzhi Jianshen Decoction", and the others were served with same dose of normal saline.After 12 weeks, general conditions of rats were observed. Then collecting 24 hours urine, to detect the 24-hour urinary protein excretion;collecting orbital venous blood, to measure blood glucose(Glu), glycosylated hemoglobin(HA1C), serum creatinine(Scr) and blood urea nitrogen(BUN). All rats were sacrificed, and the renal tissue were observed by immunohistochemical and pathological section. Levels of AGT and IGF-1 were detected by ELISA.Results(1)Compared with the blank group,the body weight of the model group and treatment group were reduced markedly(p<0.01), and ratio of kidney weight to BMI, bioch-mical indexes, levels of AGT and IGF-1 were increased significantly(p<0.01). Pathological examination of kidney in rats showed glomerular hypertrophy.mesangial matrix proliferation, glomerular basement membrane thickening, tubular epithelial cell degeneration, protein cast formation in some renal tubules.(2) Compared with the model group,the body weight of the treatment group was remarkably increased(p<0.01).,while the levels of 24 albuminuria,BUN,SCr were significant difference between the two groups(p<0.05). The pathologic changes were alleviated in the treatment group than thay of the model group.The level of AGT and IGF-1 were markedly decreased(p<0.01).Conclusions compound "Lingzhi Jianshen Decoction" can protect the kidney function and delay the development of diabetic nephropathy on the experimental DN rats; It mechanism may be association with the reducing espression of AGT and IGF-1 and inbiting the abnormal state of activated rennin angiotensin system in diabetic nephropathy,.
