| Objects: To investigate and explore the effect of traditional Chinese medicine complex prescription—Tang Ke Decoction on early.kidney pathological changes and transforming growth factor β1 s expression in experimental diabetic rat. Methods: 78 male Wistar rats were divided into seven groups randomly. Rats in one group was given common forage, rats in the other five groups were given high fat forage and were injected streptozotocin in vena caudal is once after one month to make diabetes and the blood stasis and phlegm syndromes in TCM model. Tang Ke Decoction with expelling phlegm and activating blood circulation was lavaged separatedly after success model and 4 weeks after success model. Luo Ting Xin group was control group of western medicine. Fasting blood glucose, body weight, kidney weight, rate of kidney and body weight, Urine protein quantitation in 24 hours, blood creatinine, urea nitrogen, glycosylated hemoglobin, blood fat, blood rheology and the expression of TGF—β1 mRNA in kidney tissue were detected and ultramicrostructure of kidney was observed.Results : Tang Ke Decoction can reduce the fasting blood glucose and the rate of kidney and body weight of rats; decrease the excretion of Urine protein; reduce the level of blood and urine creatinine, urea nitrogen, glycosylated hemoglobin and blood fat; restraint the excessive increase of endogenous creatinine clearance rate, lighten or delay the damages of kidney ultramicrostructure under electron microscope (lighten kidney hypertrophy and the renal glomerulus mesenterium ground substance hyperplasia and sclerosis ); restrain the expression of TGF—β1 mRNA in model kidney cortex of rats; Tang Ke prevention group had obvious curative effect compared to Tang Ke treatment group, which had significant difference.Conclusions: Chinese medicine complex prescription—Tang Ke Decoction can prevent and cure early diabetic nephropathy of rat which may have relationship with restrainting excessive expression of TGF—β1 in kidney tissue. |
