Function Of Qimuyishen Decoction In Treating Kidney Of Diabetic Rats And Its' Possible Mechanism

Abstract/Summary:
It is a pressing social and medical problem to find effective drugs for diabetic mellitus and a good approache for prevention and cure of chronic complication. Diabetic nephropathy is one of common serious small vessel complication of diabetic mellitus. It is the most reason of end—stage renal disease and diabetic mellitus death.Objective: To observe the effect of complex prescription Qimuyishen decoction on transforming growth factor -β1(TGF-β1) and connective tissue growth factor(CTGF) in renal tissue of diabetic rats and to investigate the kidney-protective effects of Qimuyishen decoction in diabetic rats and its kidney-protective mechanism.Method: Sixty famale Wistar rats ,2 months old, were randomly divided into normal control group(n=10) and diabetic mellitus models group (n=20). The diabetic mellitus model was made by a single intraperitoneal inocalation of 60 mg.kg-1 streptozotocin(STZ). After these models were successfully established, they were randomly divided into 2 groups, diabetic control group who received 20ml water for lavage and Qimuyishen decoction-treated group who received 30mg/(kg.d) Qimuyishen decoction for lavage . Eight weeks later ,the excretion of urinary protein was measured, the transforming growth factor–β1(TGF-β1)and connective tissue growth factor (CTGF) in renal tissue of diabetic rats were detected with immunohistochemistry of SP methods. Blood glucose, glycated haemoglobin Alc(HbAlc), serum creatinine, blood uria nitrogen, urinary albumin, serum cholesterol, triglyceride(TG),body weight, renal weight, renal weight / body weight ratio and creatinine clearance were measured .Result:1.After eight weeks, blood glucose, glycated haemoglobin Alc(HbAlc), serum creatinine(Scr), blood urea nitrogen, urinary albumin,serum cholesterol, triglyceride(TG),body weight, renal weight, renal weight / body weight ratio and creatinine clearance of treatment group were significantly highter in diabetic control group compared with those of control group(P<0.05)。Administration of Qimuyishen decoction for eight weeks might markedly attenuate the increased of blood glucose, glycated haemoglobin Alc(HbAlc), serum creatinine, blood uria nitrogen, urinary albumin, serum cholesterol, triglyceride(TG), body weight, renal weight, renal weight / body weight ratio and creatinine clearance of treatment group(P<0.05)2.After eight weeks , the excretion of urinary protein of diabetic group were significantly highter than in normal group(P<0.01),the excretion of urinary protein of Qimuyishen decoction-treated group were significantly lower compared with those of diabetic control group(P<0.01)。,3.The positive areas of TGF-β1 and CTGF in renal tissue of diabetic rats of diabetic rats were0.2106±0.0305和0.1965±0.0521, which were significantly highter than normal control group(P<0.01) in Qimuyishen decoction-treated group, the positive areas of TGF-β1 and CTGF in renal tissue of diabetic rats of diabetic rats were0.1345±0.0314和0.1189±0.0353, which were significantly lower than diabetic control group(P<0.01).4.The kindey morphological changes were examined by light microscopy. the renal pathological changes showed renal tubular injury ,necrosis , focal atrophy, partial tubular structure diaorder, obvious vacuole, interstitial edema and fibrosis in the test groups.Conclusions:1.The renal pathological changes showed renal tubular injury ,necrosis , focal atrophy, partial tubular structure disorder, obvious vacuole, interstitial edema and fibrosis in the test groups.2.Qimuyishen decoction-treated group can produce protectine effects on the kidney of diabetic rats .3.The kidney-protective mechanism might be that complex prescription Qimuyishen decoction down-regulates the expression of TGF-β1,CTGF which inhibits renal hypererophy and reduces syhthesis of extracellurlar matrix.
