Runchang Jiangzhi capsules is a hospital preparation of Shanghai Changzheng Hospital, processed by Rhubarb, Cassiae semen, Raphani semen and Nelumbinis folium. It is widely used for treating simple obesity, or those accompanied by hyperlipemia patients. However, the present quality standard of Runchang Jiangzhi capsules is very simple, only containing pharmaceutical properties, TLC method of Rhubarb and relative density. We improved the quality standard by adding qualitive analysis by TLC method of Cassiae semen and Nelumbinis folium, as well as simultaneous quantitative determination of aloe-emidin, rhein, emodin, chrysophanol and physcion by HPLC method. Moreover, the new quality standard of Runchang Jiangzhi capsules was drafted. It provides scientific basis for further improving the quality of Runchang Jiangzhi capsules.Fufang Shatagan Oral Liquid is also a hospital preparation of Shanghai Changzheng Hospital,processed by Chuanxiong Rhizoma and Hirudo. It is widely used for treating paralytic and hemiplegic patients. However, the present quality standard of Fufang Shatagan Oral Liquid is also very simple, only containing pharmaceutical properties, TLC method of Hirudo and relative density. To improve the quality standard, Chuanxiong Rhizoma was identified by TLC method, and the content of ferulic acid was determined by HPLC method. Moreover, the new quality standard of Fufang Shatagan Oral Liquid was drafted. It provides scientific basis for further improving the quality of Fufang Shatagan Oral Liquid.Plants of genus Siegesbeckia are annual herbs widely distributed in south of the Yangtze River and southwest of China. The Chinese Pharmacopoeia(2015 edition) has recorded the aerial parts of three species of Siegesbeckia, including Siegesbeckia orientalis L., Siegesbeckia pubescens Makino and Siegesbeckia glabrescens Makino as the traditional Chinese medicine Xi-xian to treat rheumatic arthritis,hypertension, malaria, neurasthenia, snake-bite and other diseases.This thesis investigated the chemical constituents of the aerial parts of Siegesbeckia glabrescens Makino collected from Jiujiang, Jiangxi province. Modern separation technology, such as silica gel, ODS,Sephadex LH-20 and MCI column chromatography, were applied to the isolation and purification of Et OH extract of Siegesbeckia glabrescens Makino. The structures of the compounds were elucidated by spectroscopic analysis methods, including NMR, MS and UV. At present, we have isolated and identified25 compounds from Siegesbeckia glabrescens Makino, mainly including diterpenoids, riterpenoids,steroids, flavonoids, and phenolics. The compounds successively named, n-butyl octadec-9,12-dienoate(B-01), octadeca-9,12-dienoic acid ethylester(B-02), 2-butoxyethyl linoleate(B-03), linoleic acid(B-04),n-hexadecane(B-05), stigmasterol(B-06), 4-hydroxy-3,5-dimethoxybenzaldehyde(B-07), coniferaldehyde(B-08), 4-hydroxybenzaldehyde(B-09), n-hexacos-5,8,11-trienoic acid(B-10), glyceryl-1-octadec-9',12',15'-trienoyl-2-octadec-9'',12''-dienoyl-3-hexadecanoate(B-11), ?-sitosterol(B-12), n-tetratriacont-20,23-dienoic acid(B-13), stigmasteryl-3?-arachidate(B-14), ursolic acid(B-15), ursolic acid(B-16),daucosterol(B-17), stigmasterol-3-O-glucopyranoside(B-18), ent-kaurane-16?,17-diol-19-oic acid(B-19),(2S,3S,4R,10E)-2-[(2R)-2-hydroxyl-tetracosanoyl-amino]-10-octadecene-1,3,4-triol(B-20), ent-2-oxo-15,16,19-trihydroxypimar-8(14)-ene(B-21), oleanolic acid(B-22), indole-3-carboxylic acid(B-23), ent-15-oxo-2?,16,19-trihydroxypima?-8(14)-ene(B-24), quercetin 3-methyl ether(B-25). All of the compounds are obtained from Siegesbeckia glabrescens Makino for the first time, except for compounds B-12, B-17,B-19 and B-25. The results will be supplement for the research on chemical constituents of Siegesbeckia glabrescens Makino and genus Siegesbeckia, and will lay the foundation for the subsequent study of pharmacological activity. Moreover, they are meaningful in providing scientific evidence to further development and utilization of the traditional Chinese medicine.
