FHY oral liquid is a compound traditional Chinese medicine product made by the Chinese medicine Codonopsis,Astragalus,Jujube and Glycyrrhiza after water extraction and concentration.It can mobilize the human body's disease resistance,and has the effects of strengthening the brain and nourishing blood,nourishing Qi and nourishing blood.Clinical studies of traditional Chinese medicine had shown that FHY oral liquid can be used to treat patients with insufficient vitality and immunodeficiency immediately after surgery,and has a significant effect on the treatment of deficiency and immunodeficiency.In order to further broaden the scope of application of FHY oral liquid,we had studied the protective effect of FHY oral liquid on liver damage in rats and chicks caused by AFB1,and the development of ointment formulations..The specific research content is as follows:?1?1)In this study,we established a rat model of acute liver injury induced by AFB1.By observing changes in liver index,liver tissue pathology improvement,improvement of serum liver function index,changes of immune related protein content,lipid peroxidation level of liver tissue,change of total antioxidant capacity of liver tissue,improvement of antioxidant enzyme activity,and cell color of liver tissue changes in the expression of protease were used to study the effects and possible mechanisms of FHY oral liquid on acute liver injury induced by AFB1 in rats.The research results showed that the dosage of FHY oral liquid in the range of 0.5-2 m L/kg can effectively inhibit the decline of liver index in rats caused by AFB1,improve pathological damage to liver tissue,and improve immune related factors such as ALB,TP and GLB The levels of indicators;decrease serum ALT and AST levels;significantly increase the SOD activity of liver tissue and the activity of enzymes such as CAT and GSH-Px and reduce the level of lipid peroxidation in liver tissue of rats.The results of molecularstudies showed that the expression of CYP1A2 in FHY oral liquid group of0.5-3 m L/kg was extremely significantly lower than the model group and returned to normal levels,and the inhibitory effect was dose-dependent;when the dose was 2 m L/kg,the expression of CYP3A4 was significantly lower than that of the model group.The above results suggested that:?1?0.5 m L /kg FHY oral liquid could effectively treat rat liver damage caused by AFB1,and 2 m L / kg is the best treatment;?2?FHY oral liquid was to improve the body's own immunity and improve the total liver tissue Antioxidant ability to reduce the level of lipid peroxidation in liver tissue of rats,so as to achieve the effect of treating liver damage caused by AFB1;?3?FHY oral liquid mainly treated liver damage of rats caused by AFB1 by reducing the expression of CYP1A2 in liver tissue,and decreasing the content of CYP3A4 plays a certain auxiliary role in the treatment of liver injury induced by AFB1 in rats.?2?In this study,we established a chicken liver injury model induced by AFB1.By observing changes in liver index,liver tissue pathology,improvement of serum liver function,content of immune-related proteins,lipid peroxidation of liver tissue,antioxidant activity and antioxidant enzyme activity,we investigated the therapeutic effects of FHY oral liquid on AFB1-induced chicken liver.The results showed that after feeding a feed containing 2.8 mg/kg of AFB1,the AST and ALT levels in the serum of chickens increased significantly;the ALB,TP,and GLB levels decreased significantly;the MDA content in liver tissues also increased significantly,SOD activity and CAT,GSH-PX,and GR activities were correspondingly reduced;liver histopathological damage was severe,bleeding spots were observed on the surface,and these symptoms did not improve 24 days after stopping feeding of AFB1.The above results indicated that AFB1 can cause reduced immunity,oxidative stress damage to liver tissue and the body of the chicken,and the damage lasts for a long time and is not easy to recover.After feeding 0.8-2 m L/kg FHY oral solution for 8 days,the contents of AST and ALT basically returned to normal;the contents of immune proteins such asALB,TP and GLB also increased significantly;the content of MDA in liver tissue decreased significantly,SOD activity and CAT,GSH-PX And GR activity were significantly enhanced;the pathological damage to liver tissue was also significantly restored.The results suggested that FHY oral liquid could treat chicken liver damage caused by AFB1 by improving the immunity and antioxidant capacity of chicks.?3?Using traditional paste preparation methods,the preparation process of FHY ointment was preliminarily studied.With the three indicators of moisture content,relative density,and dynamic viscosity as the detection indicators,the ointment collection steps in the paste preparation process were visualized with indicators to facilitate industrial production.The results showed that the prepared ointment has a moisture content of 25-27%,a relative density of about 1.18 g/cm3,and a viscosity of 100-300 m Pa·s.The prepared ointment has a long storage time and a moist taste,meet the requirements of ointment preparations.
