In recent years,Qingwen Jiedu Oral Liquid is more popular antipyretic and detoxifying drugs in the field of veterinary medicine.The oral liquid is a pure traditional Chinese medicine.Its efficacy is to clear away heat and detoxify toxins and to treat exogenous fever.However,as we know,The extraction rate of active ingredients from traditional Chinese medicinal materials is low,the loss of impurity removal and concentration is high,and the energy consumption is high.However,the national drug testing standards for this drug is qualitative identification and lack of the detailed composition of drug itself.This will affect the quality of drugs,even lead to clinical use of the effect.To solve the problems above,the method of high performance liquid Chromategraphy(HPLC)for the determination of forsythin,an important component of Qingwen Jiedu Oral Liquid,was established to determine the content of forsythin in the oral liquid.By studying the new preparation technology of crushing and extracting,filtering and removal of impurity,the yield of active ingredients of the oral liquid was increased,the resource wastage was reduced,and the production cost was saved.The quality of Qingwen Jiedu Oral Liquid was improved by optimizing the preparation technology.The research results of this thesis are as follows:(1)Establishment of the HPLC method for forsythinThe high performance liquid chromatographic method for the determination of forsythin was established.The chromatograph conditions are as follows:Kromasrl C18(250 mm*4.6 mm*5 um)column is used with acetonitrile and 0.2%(mass fraction)phosphoric acid solution(22:78)as mobile phase.The detection wavelength is 274 nm,the flow rate is 1.0 mL/min,the column temperature is 40?,and the injection volume is 10?L.The results showed that the experiment had high accuracy,stability and repeatability,and can be used as a quantitative detection method for forsythin.(2)The optimization of crushing and extracting parameters for Qingwen Jiedu Oral LiquidAfter the crushing pretreatment of the raw materials of Qingwen Jiedu Oral Liquid,it was found that the content of forsythiaside was different with different crushing degree.According to the single factor experiment,the results showed that the best condition is that the medicinal material is pulverized to a 24-mesh sieve.The extraction time of this process is 50 minutes.To achieve the best effect,the temperature should be 90?,and the ratio of water to medicinal material is 8:1.Under the above technological parameters,the extraction amount of active ingredients was the highest.(3)The optimization of filtration and removal of impurity parameters for Qingwen Jiedu Oral LiquidAfter crushing and extracting,the medicines were filtered by ceramic membrane.The results showed that the optimum filtration parameters of the ceramic membrane were as follows:the ceramic membrane diameter is 0.6?m?the temperature is 25?,and the pressure value is 0.20 MPa.The final product of the Qingwen Jiedu oral liquid was high in content and good in clarity.(4)pilot-scale production of Qingwen Jiedu Oral Liquid by new technology.According to the optimized technological parameters in the laboratory,Qingwen Jiedu Oral Liquid was produced in pilot scale.The test results showed that the content of active ingredients in the liquid could reach the peak value,and the extraction rate of active ingredients reached 42.8%.According to the long-term test data,in the six months of the study,the contents,properties and physical and chemical properties of forsythin were in line with the regulations.The stability of Qingwen Jiedu Oral Solution produced by the new process can be guaranteed after pilot-scale production.It is confirmed that the feasibility of the optimized process is verified.
