SnRK2(sucrose non-fermenting 1-related protein kinase 2)is a plant-specific serine / threonine protein kinase that participates in a variety of signal transduction in plants,responses to abiotic environmental stress,and some physiological process.In the previous study,we screened that the interacting protein of At SnRK2.4 was a R2R3-MYB transcription factor At MYB21.The plant R2R3-MYB protein plays an important role in plant metabolism,hormone and abiotic stress response,and plays a key role in regulating the differentiation of plant cells and the formation of tissues and organs of leaves and anthers.In this study,we proved that the full-length sequence of At MYB21 and the individual C-terminal region have transcriptional activation activity by yeast hybridization system.At MYB21 interacts with At SnRK2.4 only in the presence of both At MYB21 DNA binding domain and C-terminal transcriptional activation domain.The expression of AtSnRK2.4 and AtMYB21 in Arabidopsis thaliana flower was the highest of the tissues by real-time fluorescence quantitative PCR.Expression of AtSnRK2.4 and AtMYB21 gene were induced by NaCl,ABA and heat,and changed with the time extending.The resistance analysis of AtSnRK2.4 and AtMYB21 overexpression plants further indicated that AtSnRK2.4 and AtMYB21 were genes related to plant stress tolerance.The homozygous mutants of AtSnRK2.4 and AtMYB21 were obtained by CRISPR / Cas9 technique respectively.Which laid a theoretical and material basis for further exploring the molecular mechanism of AtSnRK2.4 and AtMYB21 in plant stress response.
