Objective:To observe Salvia injection (lyophilized) for myocardial ischemia-reperfusion injury in rat arrhythmia incidence, myocardial infarct proportion, myocardium pathological changes and influence on Cx43 and TNF-alpha, IL-6, IL-10, study on Salvia miltiorrhiza powder for myocardial ischemia-reperfusion injury in rat model of protection and mechanisms.Methods:Clean male SD rats were divided into Salvia injection (lyophilized) high dose group (250mg/kg), Salvia injection (lyophilized) low dose group (40mg/kg), Sham-operated Group (thread without ligation) and model groups. Continuous to drug 14d, through ligation the left anterior descending coronary artery 30min, again perfusion 60min, established myocardial deficiency blood again perfusion injury model, observation ECG and the rhythm disorders score, observation big rat myocardial infarction range and the myocardial organization pathology form, enzyme joint immune method detection the Group big rat serum in the TNF-alpha, and IL-6, and IL-10 content and the myocardial organization in the Cx43 of expressionResults:1.Salvia injection (lyophilized) high dose and low dose can reduce myocardial ischemia reperfusion arrhythmia of rats, reduces arrhythmia score (P<0.01 or P<0.05).2.Salvia injection (lyophilized) high dose and low dose were significantly reduced myocardial infarct size in a rat model (P<0.01 or P<0.05),the high dose group better than the low-dose group (P<0.01).3.Salvia injection (lyophilized) high dose and low dose may improve the pathologic changes of myocardial tissue.4.Myocardial ischemia-reperfusion injury rats serum TNF-α, IL-6 content compared with Sham-operated Group increased significantly (P<0.01),Salvia injection (lyophilized) high dose and low dose can be reduced serum levels of TNF-α,IL-6 (P<0.01 or P<0.05),high doses better than low dose (P<0.01);model of myocardial ischemia-reperfusion injury in rat serum IL-10 levels increased compared with Sham-operated Group (P<0.05),Salvia injection (lyophilized) high dose and low dose can increase serum IL-10 in rats (P<0.01), high doses better than low dose (P<0.05).5.Myocardial ischemia-reperfusion injury model in rat myocardial tissue expression of Cx43 and the Sham group was significantly lower (P<0.01),Salvia injection (lyophilized) high dose and low dose increased Cx43 expression in myocardial tissue of rats (P<0.01 or P<0.05), high doses better than low dose (P<0.01).Conclusion:1.Salvia injection (lyophilized) can reduces myocardial ischemia-reperfusion arrhythmia in rats, reduce the myocardial infarct proportion,improving myocardial histopathologic pattern, has protective effects on Myocardial ischemia-reperfusion;2.Salvia injection (lyophilized) can inhibit myocardial ischemia-reperfusion injury in rat serum TNF-α,IL-6 generation, increase the content of serum IL-10 and Cx43 expression in myocardial tissue, to MIRI’s protective effects may be related to it can inhibit the inflammatory response and increase Cx43 expression, high dose reduction is greater than the low-dose.
