BackgroundAcute myocardial infarction(AMI) is based on coronary atherosclerosis then occurred plaque rupture ,thrombus formed or angiospasm , which caused vascular occlusion , the cardiac muscle which the blood vessel dominated will occure durate and sever acute ischemia, then necrose the last.Clinical manifestations of the sternum after enduring severe pain, and increased serum myocardial enzyme electrocardiogram to change, which complications, high mortality, treatment on demand as soon as possible to complete re-establishment of myocardial perfusion, so we often use vein thrombolytic therapy and interventional therapy, to deoppilate obstructed blood vessel and recover the utility dabble of myocardial blood flow, rescue damaged cardiac muscle.In recent years, Chinese medicine treatment of AMI have made good effect. The effect of activiting blood circulation and removing blood stasis Salvia injection is the clinical treatment of myocardial infarction commonly used drugs.To investigate the curative effect and mechanism Salvia injection on myocardial infarction, the experiment of rat model of myocardial infarction, and Salvia injection intraperitoneal injection of 10 days to observe its experimental myocardial infarction in ST segment, serum enzymes, myocardium and hemorheology indicators impact.To supply experiment according for Salvia injection to treat myocardial infarction on clinical. Experiment 1: The effect of Salvia injection on myocardial infarction ischemic injury in ratsObjectiveTo observe the protective effect of Salvia injection on experimental myocardial infarction in rats.MethodsThe animal model: the Sprague-dawlay (SD) 32 malerats, weight (230±20) gram. Animals were randomly divided into four groups: Control group, Model group, Low molecular weight heparin Calcium injection control group (heparin group), Salvia injection group (salvia group). In order to make a rat model of experimental myocardial infarction, ligate of the left coronary artery of the rats, having been injected 45 mg/kg 3% sodium pentobarbital into abdominal, were cut off. animals were randomly divided into four groups, Blank control group (control group), model group, low molecular weight heparin calcium injection control group (heparin group), Salvia injection group (salvia group). Modeling the success of drug treatment began the next day, Salvia Group intraperitoneal inject of 1 mL Salvia injection (equivalent to the primary drug 1.5 g), intraperitoneal injection of heparin group 1 mL low-molecular-weight heparin calcium (113 IU), the control group and model group both intraperitoneal inject of 1 mL normal saline. After give the rats medication for 10 days, record the electrocardiogram, mensure the serum creatine kinase and lactate dehydrogenase , make cardiac muscle tissues HE dyeing.Results1. The effect on electrocardiogram ST segment: Compared with the control group, the ST segment of model group rats elevate obviously (P <0.01). Compared with the model group, the ST segment of the SMI group rats elevation to decreased (P <0.05), ST segment of heparin group rats decreased obviously (P <0.01). 2. The effect on serum CK and LDH: Compared with the control group, the values of serum CK and the values of LDH increased obviously in model group, Salvia groups and heparin group(P <0.01). Compared with the model group, serum CK value of Salvia group and heparin group increased (P <0.01), LDH value of Salvia group and heparin group increased obviously (P <0.05).3. The effect on morphology of myocardial tissue: The cardiac muscle textile fiber is neat, no edema, no degeneration, no necrosis and cardiac hemorrhage in control group.In modle group, endomembrane l of myocardial and papillary muscles is necrosis, there were a large number of inflammatory cell infiltration , left ventricular muscle fibers take disorder, there is edema, necrosis, even vascular necrosis and leakage of erythrocytes. Salvia groups and Heparin group shows that small pieces endomembrane of myocardial necrosis can be seen occasionally, there were a little inflammatory cell infiltration, left ventricular muscle fibers take shape disorder. Compared with modle group.the extent and scope of the edema and necrosis were less serious. Experiment 2: The effect of Salvia injection to hemorheology in rats with myocardial ischemiaObjectiveTo observe the effect of Salvia injection on myocardial infarction in rats hemorheology, to explore the mechanisms of effect on activiting blood circulation and removing blood stasis.MethodsThe modele is same as above, after treatment 10 days, from the rat carotid artery blood to detect 4 mL for hemorheology indicators.ResultsThe effect on blood rheology: Compared with the control group, whole blood high-shear viscosity, whole blood low-shear viscosity, plasma viscosity, erythrocyte aggregation index of the model group increased obviously (P <0.01), red blood cell deformability decreased obviously (P <0.01 ). Compared with the model group, the whole blood high-shear viscosity, whole blood low-shear viscosity of SMI group decreased obviously (P <0.01), plasma viscosity and erythrocyte aggregation index decreased (P <0.05), ability to enhance red blood cell deformation (P <0.05). Whole blood high-shear viscosity, whole blood low-shear viscosity, of Heparin group decreased obviously (P <0.01), plasma viscosity and erythrocyte aggregation index decreased (P <0.05), red blood cell deformability increase (P <0.05).ConclusionsSummary up, Salvia injection can promote myocardial infarction in the ST segment of the electrocardiogram recovery, to reduce myocardial infarction in the myocardial tissue edema, necrosis, the experiment shows that the Salvia injection have the protection on myocardial cells. Salvia injection can increase the serum CK, LDH level in myocardial infarction obviously, manifested that they could have a coronary artery-recanalization effect. Salvia injection can reduce the plasma, whole blood viscosity and erythrocyte aggregation index, manifested that improve hemorheology may be one of the action mechanisms of Salvia injection in myocardial protection.
