Acute myocardial infarction (AMI), commonly occurring with serious clinical consequences, often causes some myocardium suffering non-convertible injury called myocardial ischemia-reperfusion injury (I/R injury). I/R injury is also a complicated clinical pathophysiologic process, which is found during cerebral infarction, organ transplantation, etc.Myocardial I/R injury was first performed by Jennings et al. in1960in their description of the histological features of reperfused ischemic canine myocardium. Previous studies suggest that the mechanism of myocardial I/R injury consists of reactive oxygen species (ROS), Ca2+overload, neutrophil recruitment, and apoptosis etc. Neutrophils play not a commence role but a most gravity role. How to modulate the neutrophils and control the infiltration becomes the most interest issue for cardiovascular research.To investigate the cardio-protective effect of "Salvia miltiorrhiza Ligustrazine Hydrochloride and Glucose injection"(SL) induced ischemic pre-conditioning (IPC) following myocardial ischemia-reperfusion (I/R) injury in mice. We established the murine model to mimic myocardial ischemia-reperfusion injury. The proximal left anterior descending coronary artery (LAD) was subsequently ligated for30minutes for all groups, and cardiac function was monitored by echocardiography at varying time intervals (2h,8h,24h,48h,72h) following reperfusion. Cardiac function of mice in SL+I/R-20group and SL+I/R-30group were greatly improved at24and48hours following I/R. In addition, both groups displayed significantly reduced inflammation, reduced infarct size (TTC staining), decreased expression levels of TNF-a and increased expression of IL-10(Western blot). These findings reveal the possible cardio-protective effect of "Salvia miltiorrhiza Ligustrazine Hydrochloride and Glucose injection" and the reduction of I/R injury through regulation of inflammatory cytokines.
