Objective:To observe the effect of injection with salvia miltiorrhiza(lyophilization)on whole blood viscosity,plasma viscosity,concentration of NO and H2O2 in cardiomyocytes,the expression of PMCA,SERCA and Ca2+-magnesium2+ATPase proteins in cardiomyocytes of rats with myocardial ischemia/reper fusion injury.To investigate the myocardial protective effect and its mechanism of injection with salvia miltiorrhiza(lyophilization)on myocardial ischemia/reperfusion injury in rats.Methods:Clean male SD rats were randomly divided into injection with salvia miltiorrhiza(lyophilization)high dose group(250mg/kg),injection with salvia miltiorrhiza(lyophilization)low dose group(40mg/kg),model group and sham operation group.Two weeks of continuous administration,through the ligation of the left anterior descending coronary artery 30min,reperfusion 60min,establish the model of myocardial ischemia/reperfusion injury in rats.Whole blood viscosity and plasma viscosity were measured by anticoagulation,concentration of NO and H2O2 in cardiomyocytes was detected by myocardial tissue and the expression of PMCA,SERCA and Ca2+-magnesium2+ATPase proteins in cardiomyocytes by Western blotting.Results:1.Compared with the model group,the injection with salvia miltiorrhiza(lyophilization)high and low dose group can significantly reduce the shear rate of the whole blood viscosity and plasma viscosity(p<0.01 or p<0.05),high dose group is better than the low dose group.2.Compared with the model group,the injection with salvia miltiorrhiza(lyophilization)high and low dose group can significantly reduce the concentration of NO and H2O2 in myocardial cells(p<0.01),high dose group is better than the low dose group.3.Compared with the model group,the injection with salvia miltiorrhiza(lyophilization)high and low dose group can significantly increase the protein expression of SERCA and Ca2+-Mg2+-ATPase in myocardial cells(p<0.01 or p<0.05),high dose group is better than the low dose group.Conclusions:1.Injection with salvia miltiorrhiza(lyophilization)can significantly reduce the whole blood viscosity and plasma viscosity of MIRI rats,the protective effects on MIRI rats may be related to the improvement of blood circulation and blood viscosity.2.Injection with salvia miltiorrhiza(lyophilization)can significantly reduce the concentration of NO and H2O2 in myocardial cells of MIRI rats,the protective effects on MIRI rats may be related to the reduction of oxidative damage,the improvement of antioxidant capacity and the inhibition of apoptosis in cardiomyocytes.3.Injection with salvia miltiorrhiza(lyophilization)can significantly increase the expression of SERCA and Ca2+-Mg2+-ATPase proteins in MIRI rats cardiomyocytes,the protective effects on MIRI rats may be related to the reduction of intracellular calcium overload.
