| Objective:To investigate the potential association between the OPN gene polymorphismsand idiopathic calcium oxalate stones in Jiangxi Gannan area.Methods:We collected the blood and urinary samples from218patients with ICS (ICSgroup) and187normal people (control group) in Jiangxi Gannan area. The factorsaffecting the formation of ICS were analyzed by the logistic regression analysis, suchas age, sex, body mass index (BMI), urine and blood biochemical indexes. OPN genepolymorphisms were detected by real-time fluorescent quantitative polymerase chainreaction TaqMan-MGB5’-nuclease assay. The correlation between the OPNpolymorphisms and ICS was analyzed.Results:Single factor analysis showed that BMI and urine calcium concentration weresignificantly higher in ICS patients than that in controls, and the PH values of urineand fluid intake was lower in the ICS patients compared with controls (P<0.05).Logistic regression analysis showed that the urine calcium concentration was stillstatistically significantly different between the two groups (P=0.000). Thedistribution frequencies of three SNPs genotype in the two groups followed theHardy-Weinberg Equilibrium (P>0.05). The distribution frequencies of the delG andG alleles of rs11439060in ICS and control group were69%,31%and57%,43%,respectively (P=0.000). The frequency of the delGdelG, delGG and GG genotypewere45.9%,46.3%and7.8%in ICS group, and32.1%,49.7%and18.2%in controlgroup. The risk for ICS of delGdelG and delGG genotype were3.333and2.172timesthat of GG genotype (P=0.000, OR=3.333,95%CI=1.715~6.478and P=0.017,OR=2.172,95%CI=1.138~4.147, respectively). The frequency of the C and T alleles of rs6840362in ICS and control group were91.7%,8.3%and95.7%,4.3%,respectively (P=0.021). The frequency of the A and G alleles of rs6532040in ICS andcontrol group were90.1%,9.9%and94.4%,5.6%, respectively (P=0.026). There wasno statistically significant difference between the two groups with the genotypesfrequency for rs6840362and rs6532040(P>0.05). There was no statisticallysignificant difference in the relations between the genotypes and clinical phenotype(P>0.05). OPN gene haplotype analysis showed that haplotype C-G-delG waspositively correlated with the formation of ICS (OR=6.326,95%CI=1.958~20.442),but haplotype C-A-G was negatively correlated with the formation of the ICS(OR=0.600,95%CI=0.448~0.804).Conclusions:1. OPN gene polymorphisms were significantly associated with the formationof ICS in Jiangxi Gannan area. In rs11439060, genotype delGdelG and delGG andhaplotype C-G-delG may be the risk factors for the formation of ICS, but genotypeGG and haplotype C-A-G may be decreased the formation of ICS.2. Increased urine calcium concentration may be a risk factor for the ICSformation. |
