Objective:To research the changing law of IL-8,IL-10 level before and after the fat-loading and its relationship with atherosclerosis.Methods:Selected 10 healthy Male New Zealand white rabbit,only 2 to 3 months,2.5-3 kg,were given the load high fat diet(1% cholesterol,8% egg yolks, 91% basic feed) for 12 weeks.On initial experiment and the 4 th, 8 th, 12 th week, all rabbits were given a fat meal load(2%cholesterol), prior to the load and the load respectively after 2 th, 4 th, 6 th, 8 th hour ear marginal vein blood determination of serum IL-8 and IL-10 level. Over the same period,ultrasonic observation of abdominal aorta situation and ultrasound scores. At the end of the experiment, put to death all the experimental animals, observed animal abdominal aortic sclerosis under electron microscope with HE staining.Results:1. The level of IL-8 changes before and after high cholesterol load:In the process of experiment, along with the hardening of the arteries the level of IL-8increased on an empty stomach(P <0.05), 4 th week up to the peak(P <0.05) and 8 th,12 th week gradually decrease(P >0.05). It was a little high at the 2 th to 6th hour pro the meal on the initial(the highest 6 th hour, P <0.05); It was higher at each time point pro the meal than the fasting on the 4 th and the 8 th week(the highest 4th, 6 th hour, P <0.05); At the end of experiment,there was no obvious change of the level of IL-8 pre and pro the fat-loading(P >0.05).2. The level of IL-10 changes before and after high cholesterol load:Along with the hardening of the arteries progress,the levels of IL-10 were higher on an empty stomach, a significant rise on the 4 th and 8 th week(P < 0.05), 4 th week is highest and 8 th,12 th week gradually decline.The levels of IL-10 were slightly higher after the meal on initial experiment and 4 th week, 4 th hour after the fat-loading most obviously(P <0.05);But it’s level was not apparent changed pre and pro the fat-loading on the 8th week(P >0.05); At the 12 thweek, the levels of IL-10 were first increased and then reduced,It was slightly higher at the 2 th,4th hour(P <0.05),and was slightly lower at the 8th hour after the meal(P <0.05).3. In the process of the experimental, the correlation of IL-8 and IL-10 before and after fat meal loading:Fasting state, there was a reverse correlation on the initial experiment and 8 th,12 th week, and a positive correlation on the 4thweek. After the fatty meal load, there was a positive correlation at the 2 th hour, the rest was a reverse correlation on the initial experiment;On the 4thweek, 8thhour was a positive correlation, the rest was a reverse correlation; On the 8 th week, there was a positive correlation at the 6 th hour, the rest was a reverse correlation; On the 12 thweek, pre and pro the fat-loading are negatively correlated,especially at the the fasting time and 6 th hour.4. In the process of hardening of the arteries, changes of IL-8/IL-10 pre and pro the fat-loading:During the experiment, The IL-8/IL-10 ratio was slightly increased on the 4thweek(P<0.05);There was no obvious change at each experiment time point pre and pro the meal on the initial and the 8th, 12thweek(P >0.05).5. Experimental animal abdominal aorta ultrasound examination:On the initial, all animal abdominal aorta wall smooth, not coarse; Animal experiments for 4 th week, walls are rough, and visible-membrane in varying degrees of thickening; For8 thweek, 6 abdominal aorta wall of tiny patches of experimental animals, the rest of were obvious thickening; For 12 th week, all animal abdominal aorta plaque formation.If the experiment in the study the initial abdominal aorta ultrasonic score is defined as1, Found: progress along with the hardening of the arteries, abdominal aorta ultrasonic experimental animals integral rise gradually, and observe the following conditions:(1) the intima rough, not smooth;(2) the IMT thickened gradually;(3) at the end of the study,abdominal aorta wall are visible to the patches.6. The correlation of IL-8 and the AS ultrasonic score before and after the high cholesterol loading:(1) In the experiment of the 4thweek, 4thand 6thhour after the high cholesterol loading was negative correlation,the rest was a positive correlation(2 th hour best);(2) In the experiment of the 8thweek, 4thhour was positive correlation after the high cholesterol loading, the rest was a negative correlation(2thhour best);(3) In the experiment of the 12 th week, 4thhour was positive correlation after the high cholesterol loading, the rest was a negative correlation(especially at the the fasting time).7. The correlation of IL-10 and the AS ultrasonic score before and after the high cholesterol loading:(1) In the experiment of the 4thweek, there was a positive correlation on an empty stomach and each time point after the high cholesterol loading(especially at the 4th and 6th hour);(2) In the experiment of the 8thweek, 6thhour was negative correlation after the high cholesterol loading,the rest was positively correlated(2 th hour best);(3) In the experiment of the 12 thweek,there was a positive correlation at the fasting and each time point after the high cholesterol loading(especially at the 2th hour).8. Experimental animal abdominal aorta macroscopic observation and pathological examination:macroscopic observation:At the end of the experiment(12thweeks), all the animals’ abdominal aorta profile could observed the plaque.pathologic examination:Normal blood vessels: endothelium is integrated, smooth muscle cells arranged orderly, without foam cells; At the end of the experiment, abdominal aorta vascular endothelial cell swelling, intima rough and damaged, Smooth muscle cells arranged disorder, along with a large number of foam cells and obvious "empty pulp phenomenon".Conclusion:1. In the process of atherosclerosis, IL-8 and IL-10 mainly involved in hardening of the arteries of the early inflammatory reaction process. In the early atherosclerosis,it exists the imbalances of proinflammatory factor and anti-inflammatory factor.2. The experiment shows that the fat-loading had a greater influence on the early inflammatory response to arteriosclerosis. However, in the later of hardening of the arteries,the body anti-inflammatory ability enhancement after the fat-loading.
