Objective:Research changes of advanced glycation end products(AGEs)and vulcanized dehydroepiandrosterone(DHEAS)before and after the fat-loading and its relationship with atherosclerosis.Methods:Select 10 healthy male New Zealand white rabbits(3 mouths ago,weighing 3.0±0.25kg),high-fat continuously feeding 12 weeks,high-fat feed consists of 89% basal diet,10% egg yolk,1% cholesterol.At the start and the 4th,8th,12 th week of the experiment,give them fat meal load(containing 78% basal diet,20% egg yolk,2% cholesterol),at the same time,collecting the venous blood from the ear marginal vein at fasting and 2,4,6,and 8h after fat meal load.At the same time,the abdominal aortic lesions were observed and scored.At the end of the experiment,separating the abdominal aorta and making pathological specimen to observe the ATH conditions.Results:1.Changes of plasma AGEs pre and pro the fat-loadingAfter fat meal,AGEs increased gradually at the beginning of the study,and the 8h was highest(P<0.05),but at the 4th,8th,12 th week,AGEs increased first and decreased then,the 2h was highest(P<0.05),indicating that oxidative stress was severe and clearly observed at 2h after fat meal load.With the progression of arteriosclerosis,the fasting AGEs increased gradually(P<0.05),and the highest at the 12 th week,indicating that oxidative stress gradually aggravate in the progression of atherosclerosis.2.Changes of plasma DHEAS pre and pro the fat-loadingAfter fat meal,DHEAS decreased gradually at the beginning of the study,and the 8h was lowest(P<0.05),but at the 4th,8th,12 th week,DHEAS decreased first and increased then,the 2h was lowest(P<0.05),indicating that antioxidant ability was significantly reduced at 2h after fat meal load.With the progression of arteriosclerosis,the fasting DHEAS decreased gradually(P<0.05),and the lowest at the 12 th week,indicating that antioxidant capacity was gradually decreased in the progression of atherosclerosis.3.Ultrasound observation of abdominal aortaWith a long period of high fat feeding,the abdominal aorta of rabbits change from smooth lining to rough,and the IMT thickened gradually,the small plaques could be observed at 8th week,and at the 12 th week was significant.4.The relationship of AGEs change and arteriosclerosis(ultrasound score)pre and pro fat meal(1)At the 4th week,postprandial 4h was negative correlation,the rest was positive correlation(significantly at fasting and the 2h)(2)At the 8th week,postprandial 4h was negative correlation,the rest was positive correlation(significantly at fasting)(3)At the 12 th week,postprandial 6h was negative correlation,the rest was positive correlation(significantly at the 2h)5.The relationship of DHEAS change and arteriosclerosis(ultrasound score)pre and pro fat meal(1)At the 4th week,postprandial 4h was positive correlation,the rest was negative correlation(significantly at the 2h)(2)At the 8th week,postprandial 6h was positive correlation,the rest was negative correlation(significantly at the 4h)(3)At the 12 th week,postprandial 6h was positive correlation,the rest was negative correlation(significantly at fasting and the 2h)6.The macroscopic observation of experimental animal's abdominal aortaAt end of the experiment,the abdominal aorta of rabbits showed poor elasticity,abnormally rough wall,and obvious plaque.7.abdominal aortic pathologyAt the end of the study,the vessel wall damaged apparently,and there were a large number of foam cells.Most of the endometrial thickened,and the endothelial cells and smooth muscle cell proliferation.Conclusion:1.At any time,the fat-loading will cause AGEs increased and DHEAS reduced(2h),suggesting that oxidative stress strengthened and antioxidant ability weakened during the early stage after fat-loading.2.During the procession of atherosclerosis,the level of fasting AGEs increasesd markedly,the level of fasting DHEAS reduced gradually,suggesting that persistent hign fat diet will aggravate the oxidative stress and weaken the antioxidant ability.
