Objective:The changes of monocyte chemoattractant protein-1(MCP-1)and adiponectin(APN)at various time points before and after the fat-loading and their relationship with atherosclerosis was studied.Methods:Select 10 Healthy Male New Zealand White Rabbits weighing 2.8±0.2 Kg,were given the high-fat diet(the composition consists of 1% cholesterol,10% egg yolk,and89% normal feed)continuous 12 th week.At the beginning of feeding,4th week,8th week,and 12 th week,all rabbits were given the fat-loading(the composition consists of 2% cholesterol,20% egg yolk,and 78% normal feed)respectively,at the same time,through ear marginal vein blood determination collect blood samples at2 th,4th,6th and 8th hours after fasting and lipid meal.After centrifugation at high speed,take the supernatant serum for cryopreservation.The concentration of MCP-1and APN was detected by ELISA method,and the changes of MCP-1 and APN were analyzed.And,the changes of rabbit abdominal aorta were observed by ultrasound.At the end of sampling,use syringe extracting air,and through the rabbit ear vein inject air kill them.The abdominal aorta was then isolated and stained with HE.The pathological changes were observed under a microscope.Results:1.MCP-1 content in the fasting state and after the change of load fat meal:With the continuous feeding of high-fat diet,the progression of arteriosclerosis in rabbits was progressed,at each time point in the fasting state,MCP-1 content was gradually increasing tendency,and the content of MCP-1 increased to its highest level at 12 th week(P <0.05).After the fat-loading,the content of MCP-1 was gradually increased at 4th week,and the content of MCP-1 was higher than that at the initial time after meal(P<0.05)and at 8th hour after meal the content of MCP-1 was highest(P<0.05);at 8th week the content of MCP-1 was highest at the 8th hour after meal(P>0.05).At 12 th week,the content of MCP-1 was highest at the 2th hour after meal(P>0.05),and then decreased gradually.2.APN content in the fasting state and after the change of load fat meal:With the continuous feeding of high-fat diet,the progression of arteriosclerosis in rabbits was progressed,at each time point in the fasting state,the content of APN gradually decreased,and the content of APN was lower than that in the initial state at4 th week,8th week,and 12 th week(P<0.05),and the APN content was reduced to the lowest at 12 th week(P<0.05).After the fat-loading,at the beginning,4th week,and 8th week the level of APN decreased first and then increased.At 2th(P>0.05),4th,6th,and8 th hours,the APN after meal was lower than the initial level(P<0.05),and the content was lowest at 6th hours after meal(P<0.05).At 12 th week,the content of APN showed a decrease-elevation-decline trend,and the lowest at 8th hours after meal(P<0.05).3.Ultrasound observation of abdominal aorta at different times:With the prolongation of time for high fat diet feeding New Zealand white rabbits,the level of arteriosclerosis in rabbits gradually intensified,and under ultrasoundv observed the extent of abdominal aortic lesions was gradually progressing.At the beginning of the experiment,the abdominal wall of the rabbit's abdominal aorta was smooth and the thickness of the wall was within the normal range.At the 4th and 8th week,the abdominal aorta intima was thicker than the initial thickness,and the wall of the rabbit became more rougher.At 12 th week,the abdominal aortic wall of the rabbit was thickened,the lumen stenosis was obvious,which is visible on the wall of the rabbit is plaque.4.The relationship between changes in MCP-1 at various time points and AS(ultrasound scores)during AS:(1)The 4th week of the experiment,it is positively correlated with postprandial MCP-1 content,and 8th hours after meal is most obvious;(2)The 8th week of the experiment,it is positively correlated with postprandial MCP-1 content,and 8rh hours after meal is most obvious;(3)The 12 th week of the experiment,8th hour is negative correlated after postprandial,the rest were positively correlation.5.The relationship between changes in APN at various time points and AS(ultrasound scores)during AS:(1)The 4th week of the experiment,it is negatively correlated with postprandial APN content,and 8th hours after meal is most obvious;(2)The 8th week of the experiment,it is negatively correlated with postprandial APN content,and 6th hours after meal is most obvious;(3)The 12 th week of the experiment,8th hour is positive correlated after postprandial,the rest were negatively correlation.6.Visual observation of changes in abdominal aorta:At the end of the experiment,the abdominal aorta of New Zealand white rabbits showed poor elasticity,abnormally rough wall,and there are obvious plaque attached to the wall.7.Pathological changes of abdominal aorta:At the beginning of the experiment,the rabbit's abdominal aortic intima was was thin and smooth,and the smooth muscle cells were arranged were arranged in order,showing a long strip.At the end of the experiment,the rabbit intima of the abdominal aorta thickening is significant,the integrity and continuity was destroyed,and forms fissures.And the vascular smooth muscle cells are miscellaneous and disorganized,and what appear under the intima is a large number of elongated or oval foam cells.Conclusion:1.In the early stage of arteriosclerosis,after the fat-loading,the inflammation is constantly increasing and the inflammatory response was the strongest after 8th hours;however,the body's anti-inflammatory capacity is constantly weakened and the anti-inflammatory capacity is lowest in 6th-8th hours.2.During the progression of AS,the inflammation is constantly increasing,the anti-inflammatory ability gradually weaken;in the middle and late stages of arteriosclerosis,the body's inflammatory response is the strongest and its anti-inflammatory capacity is the weakest.
