The Research On The Changes Of PDGF-BB?TGF-?₁ Level Pre And Pro The Fat-loading,on The Relationship With Atherosclerosis

Objective:To study the regultion of PDGF-BB?TGF-?1 before and after the Ch load and its realitionship with AS.Methods:Chose 10 healthy newland rabbit,male,2.9�0.4kg,to give this 10 rabbits Ch(1%Ch)load for 12 weeks,morever,give Ch load(2%Ch)at the begining and the fourth ?eigthth?tweenth week.at the meantime,the the blood of vessels at the time of before and after the Ch load(2th?4th?6th?8th),use the B-ultrsond to detetive the sutition of the atery at the time of before and after the Ch load,and use the chin to evaluate the sutition of the atery.AT the end of the experiment,killl the animal,take the artery from it,colour it,evaluate the sutition of the artery.Results:1.The concentration of PDGF-BB before and after the Ch load :During the period of AS,before the TC load,the highest concention of PDGF-BB is in the fourth week.on the sutition of Ch load,at the ime of before and the fourth Ch load,its concention of PDGF-BB gradually increases,at the time of eigthth week reach the highest(P < 0.05).at the eigthth week,at the time of after the meal(2th?4th)decreases compared with the concentration before the load,at the time of after the meal(6th?8th))decreases compared with the concentration before the load.at the 12 th week,the concentration of PDGF-BB decreases on the whole after the load.it suggests that the increase of SMC increases at the earily period of AS.2.The concentration of TGF-?1 before and after the Ch load:During the period of AS,before the Ch load,the highest concention of TGF-?1 is in the eigthth week,after that,the concentration of TGF-?1 decreases.on the sutition of Ch load,at the time of before and the fourth Ch load,its concention of TGF-?1gradually increases at the time of the fourth week and the eigthth week,it reach the highest at the time of 4-8th hour(P < 0.05).at the eigthth and tweenth week,at the time of after the meal,decreases compared with the concentration before the load,at the time of after the meal(6th?8th)its concention of TGF-?1 decreases compared with the concentration before the load.3.The ratio of the PDGF-BB/TGF-?1 before and after the Ch load:During the period of AS,before the Ch load,the highest concentration of PDGF-BB/TGF-?1 is in the eigthth week.on the sutition of Ch load,at the time of the fourth week,the ratio of the PDGF-BB/TGF-?1 increases and reach the top at the time of 8th hour.at the time of the eigthth week,the ratio of the PDGF-BB/TGF-?1decreases and reach the lowest at the time of 6th hour.at the time of the tweenth week,the ratio of the PDGF-BB/TGF-?1 shows no change..4.The ultrasound test of artery :At the begining of the study,the inner surface is smooth,and its width is normal.at the time of the fourth week,we can see its width increases to different extent,and the continuation broke.at the time of the eigthth week,we can see small.plaque on ith surface,but not obviously.at the end of the study,we can see obvious increation of width and obvious plaque.5.The realitionship between the concentration of PDGF-BB before and after the Ch load and its ultrasound score:(1)At the time of the fourth week,the realitionship at the time of 6th hour after the Ch load is negative,the rest is positive,4th hour is most obviously;(2)At the time of the eigthth week,the realitionship at the time of 6th hour after the Ch load is negative,the rest is positive,8th hour is most obviously;(3)At the time of the tweenth week,the realitionship at the time of 6th hour after the TC load is negative,the rest is positive,4th hour is most obviously.6.The realitionship between the concentration of TGF-?1 before and after the Ch load and its ultrasound score:(1)At the time of the fourth week,the realitionship at the time of 2th and 6th hour after the Ch load is positive,the rest is negative,4th hour is most obviously;(2)At the time of the eigthth week,the realitionship at the time of 8th hour after the Ch load is negative,the rest is positive,4th hour is most obviously;(3)At the time of the tweenth week,the realitionship at the time of 8th hour after the Ch load and before the load is positive,the rest is negative,2th hour is most obviously.7.The normal obversation of the artery in our eyes and under the microscope:At the end of the study,we can see extraordinary plaque on the surface of artery in amolst all the animals.8.The pathological test of artery At the begining of the test,the inner surface of artery is normal,and the SMC is ranked orderly;at the end of study,.the inner surface of artery is rough,and the continuation is broke,the number of SMC increases damatically and ranked disorderly,the structure of artery is not very tight,we can see a lot of empty pulp phenomenon.Conclusion:1.During the stage of AS,the PDGF-BB and TGF-?1 take part in the earily SMC proliferation reaction,and the concentration of PDGF-BB and TGF-?1 shows mutual balance.2.At the early stage of AS,the Ch load could posssibly strengthen the ability of SMC proliferation reaction,At the late stage of AS,the ability of anti-SMC proliferation reaction decreases after the fat-loading.
