Objective : Jianpi Shugan Jiangzhi Decoction is an experienced prescription summarized by tutor Lu Baoping according to his abundant clinical experiences, and it is combined by ancient traditional Chinese medicine(TCM)and modern western thoughts. It has notable curative effects in clinical application for curing of nonalcoholic fatty liver disease(NAFLD).The objective of this study is to observe the effects of traditional Chinese medicine(TCM)on the NAFLD model mice induced by high-fat diets and of drugs together, to clarify the pharmacological effects of the prescription, to explore the mechanism of the Decoction and morover to provide a theoretical basis for further research and clinical application.Methods:Take 72 of SPF Kunming mice, and they were all feed by ordinary SPF forage for 7 days, they were randomly divided into 6 groups, each group included of 6 pairs. The groups are the normal group, model control group, shell fat capsule group, three treatment groups classified by high, medium, and low dosage.Normal group was feed by SPF ordinary forage, other groups feed by high fat forage and drugs to induce NAFLD model mice.Then except for the normal group and model control group, and the normal group and model control group feed by Medical distilled water with the same volume.In addition to the normal group, each group was given the subcutaneous injection of 10% CCl4 cooking oil solution with a dosage of o.1m L/10 g, 2 times per week in the lower abdomen.The experiment lasts for 6 weeks, and fasted 12 hours after the last treatment, the mice were given intraperitoneal injection anesthesia with 10% chloral hydrate solution with the dosage of 4 m L/kg. Then took blood from abdominal aorta and had centrifugal separation in low temperature. The content of TG, TC, ALT, AST, HDL and LDL levels in serum or reactive changes and FFA were detected.After the mice were killed, weighed rapidly dissected liver and calculated the liver index. Then the livers were washed with 0.9% Na Cl solution, in the left lobe of the liver at 0.5cm from the edge cutting a small piece of a suitable size of liver tissue in 4% paraformaldehyde liquid fixed, histopathological changes were observed in liver embedded in paraffin, using HE staining.Results: ①compared with normal group, the index of mice liver, liver wet weight and serum ALT, AST, TC, LDL and FFA of model group had an obvious rise, had obvious statistical significance(P<0.01). ②Compared with normal control group, the treatment of high, middle dose group of liver index, liver wet weight, TG, TC, ALT, AST, LDL and FFA had no statistical significance(P>0.05); Treatment of low dose group and the shell fat capsule group index was statistically significant(P<0.05). ③Compared with model group, the the indicators of treatment with high, medium and low dosage group, shell fat capsule group are significantly reduced or mitigated, was statistically significant(P<0.05). ④In the aspect of improving the indicators, the group with high and medium group was superior to shell l fat capsule group and low dosage group, which had no statistical significance(P>0.05). ⑤Visible light: lobular structure was clear, around the central vein of liver cells arranged in the form of radiation with uniform morphology and size. No fatty degeneration and necrosis, inflammatory cells invasion discovered; Microscopically, structure of hepatic lobule of the model control group mice was not clear, hepatic lobule structure was obviously damaged, liver cell volume obviously swelling, and liver cells edema.Diffuse fatty degeneration of the hepatic tissues and irregular arrangement of the cells, with the increase of the nuclear cells within a single large lipid droplets to the nucleus to the cell membrane, Sometimes visible by more than 2 and 2 ruptured liver cell fusion, a "balloon" change, and the lesions near the central vein, Portal area, lobular within a large number of inflammatory cells invasion, sometimes visible apoptotic body, the treatment group and mouse hepatocyte fatty change shell fat capsule group all have different degrees of improvement trend, Treatment of high, middle dose group and the shell grease capsule group better than low dose group. ⑥ Under the electron microscope visible: normal Group liver tissue ultrastructure of liver cell has a wealth of mitochondria, mitochondrial cristae is relatively abundant, but also there are few lipid droplets, visible in the model group mice liver tissue ultrastructure of liver cell has a wealth of mitochondria, mitochondrial cristae almost disappeared, but also can be seen there are a number of lipid droplets, and a number of collagen fibers, the treatment group and mouse hepatocyte fatty change shell fat capsule group all have different degrees of improvement, treatment of high, middle dose group and the shell grease capsule group better than low dose group.Conclusion:Decoction of Jianpi Shugan Jiangzhi had good effects on the liver function, blood lipid, TG, TC, HDL and LDL liver tissue of NAFLD mice, this can reduce serum transaminase activity by facilitating the recovery of NAFLD liver cells to reduces the amount of blood lipid and the activity of FFA, narrow NAFLD liver shape and weight; Restore the mitochondria in liver cells, strengthen the liver cells and increase the role of mitochondrial cristae mitochondria, make liver cells to produce a large amount of ATP to enhance its energy metabolism, thus boosting the fatty acid metabolism, reduce the formation of lipid droplets; For NAFLD mice, The curative efficacy of three groups with high,medium and low dosage had visible difference, with the increase of dosage making more positive curative effects.
