Gompare Study Of New Xuefuzhuyu De-coction And Xuefuzhuyu Decoction On Levels Of PAF、PF4 In Atherosclero-sis Rats

Objective: By animal experiments, from the effect of platelet aggregation, to explore the difference of new Xuefuzhuyu Decoction and Xuefuzhuyu Decoction effect, provide more basis for the clinical use of new Xuefuzhuyu Decoction.Methods: 40 male Wistar rats, 10 rats in each group, were divided into 4 groups: blank group, model group, Xuefuzhuyu Decoction group and new Xuefuzhuyu Decoction group. The blank group was fed with common forage, other groups were given high fat diet. At the first, 4, 10 day of the experiment, besides blank group,other rats were injected Vitamin D3 Injection, the blank group was given an equal volume of saline by intraperitoneal injection. At the first day,rats were given the traditional Chinese medicine, Xuefuzhuyu Decoction group were given with Xuefuzhuyu Decoction 10ml/Kg;the new Xuefuzhuyu Decoction group were received new Xuefuzhuyu Decoction 10ml/kg; the rats of blank group were received gastric perfusion of distilled water 10ml/kg. All rats were weighed once a week, in order to the adjustment of the use of Chinese medicine. A total time of 8 weeks of the experiment. At the end of 8th week,,all rats were given no water and no food.Use 10% chloral hydrate intraperitoneal injection of 4.5ml/kg anesthesia in all rats, 5ml of blood in the aorta, and set aside for 30 minutes, after centrifugation, using enzyme-linked method to measure the serum PAF, PF4 content in rats. Then took the thoracic aorta of rats, washing, soaking, fixation, staining and other treatment, aortic changes were observed under light microscope, and measured the intimal thickness.Results: The PAF,PF4 content of Xuefuzhuyu Decoction group and new Xuefuzhuyu Decoction group were lower than those in the model group, P<0.05; on the content of rat serum PAF, the new Xuefuzhuyu Decoction group was lower than that of Xuefuzhuyu Decoction group, P<0.05;on the content of rat serum PF4, new Xuefuzhuyu Decoction group was lower than the blood of Xuefuzhuyu Decoction group, P>0.05. Compared with the blank group, the aortic intimal thickness of model group increased significantly, P<0.05.Compared with the model group, the Xuefuzhuyu Decoction group,new Xuefuzhuyu Decoction group aortic intimal thickness decreased, P<0.05.And compared with the Xuefuzhuyu Decoction group, the new Xuefuzhuyu Decoction group aortic intimal thickness was decreased, P>0.05.Conclusion: Xuefuzhuyu Decoction, new Xuefuzhuyu Decoction can decrease the level of serum PAF, PF4 content of arteriosclerosis in rats and inhibit platelet aggregation, by the inhibition of PAF,the new Xuefuzhuyu Decoction is better than that of Xuefuzhuyu Decoction, new Xuefuzhuyu Decoction on inhibiting platelet aggregation may be more advantages.
