| Objective:This article discusses the regulation of Rhodiola extract, Acanthopanax extract and Rhodiola-Acanthopanax extract on liver fibrosis.Methods:. Extraction with refluxing 70% ethanol.80℃, extracted 2 times(first liquid ratio:1:10;second liquid ratio:1:8),1.5 h/time. Effects of three extracts (1280ã€640〠320ã€160.80ã€40ã€20ã€10ã€0 ug/mL) on t-HSC cell viability by MTT. Administration concentration is 80ug/mL-.160ug/mL, assessment t-HSC cell expression of Bax,Bcl-2,XIAP,FLIPL,Caspas-3,Caspase-8,Csapase-9 by Western Blotting. Country of liver fibrosis model by Intraperitoneal injection of TAA (first week:100mg/Kg,3 times;four weeks later:200mg/Kg,3 times/week; total five weeks).Ig extract (100,200,300mg/kg) 5 weeks,1 time/day. Collecting serum and liver. Determination of ALT, AST activity and liver mass index. The protective effects of three extracts on liver fibrosis by histopathology.Results:Extraction rate of Rhodiola,Acanthopanax, Rhodiola-Acanthopanax is 5.12%,2.64%,5.51%. MTT results showed that three extracts can inhibit the proliferation of t-HSC cells.80-160ug/mL Rhodiola-Acanthopanax extract inhibit the proliferation of t-HSC cells significantly. Western Blotting results showed that Rhodiola-Acanthopanax extract can inhibit the expression of Bcl-2,XIAP effectively,and promote increased vigor of Bax,FLIPL,Caspase-3,8,9. We established mouse model of liver fibrosis with TAA successfully. Rhodiola-Acanthopanax extract can inhibit ALT,AST in serum and reduced liver mass index.CONCLUSION:. By experiments in vivo and vitro, Three extracts have inhibition of t-HSC cell proliferation,and have a good protective effect on TAA-induced liver fibrosis.
