Objective: To study the therapeutic effect of Rhodiola sachalin guanine on experimental rat liver fibrosis.Methods:70 healthy male SD rats were randomly selected as blank control group,the remaining 54 rats were used as liver fibrosis animal model establishment,placed in the same environment.The rats were sacrificed with CCl4 at a dose of 1.0 ml / Kg ·BW.The model of liver fibrosis was established.The control group was given normal saline at a dose of 1.0 ml / Kg · BW for a relatively fixed time each day.once a day.After 60 days of intragastric administration,the rats were randomly divided into control group.The rats in the model group were divided into three groups.The rats in the model group were treated with 0.5% chloral hydrate and the blood of the celiac arteries was collected.Detection of the indicators.After routine pathology examination,according to Table 2.2 to determine whether the model is successful.After the model was successfully established,the liver fibrosis model group was randomly divided into model group A: normal saline group 13,model group B:guanine low dose group 13,model group C: guanine high dose group 12,A,B,and C were given normal saline 1.0 ml / Kg · BW,low dose guanine 0.056 g / Kg · BW,0.112 g / Kg · BW high dose guanine,and the control group was also taken as the blank control group Of the animals were given normal saline 1.0 ml / Kg · BW.After60 consecutive days,each group of animals fasting can not help but overnight,the next day to 0.5% chloral hydrate subcutaneous injection of anesthesia after abdominal arterial blood,serum separation of liver fibrosis after the detection.The liver was fixed with 10% formalin solution and immunohistochemically examined.Through the serum index detection and pathological observation,to determine the efficacy of drugs.Results:At the end of the 60-day model,11 rats died,including blank control group 2,the establishment of liver fibrosis model group 9,the cause of death due to improper operation and carbon tetrachloride poisoning and death.(P <0.05).There were no significant differences in ALT and AST between the model group and the blank control group(P <0.05).There was no significant difference between the two groups(P> 0.05)difference.Combined with pathology and biochemical results show:modeling success.A total of 9 rats were sacrificed after 60 days of treatment.There were 1 blank control group,4 rats in the model group,3 in the model group,only 3 in the model group and 1 in the model group.Drug operation improper death.Biochemical indicators showed: ALT,CIV,LN were statistically significant(p <0.05).AST was not statistically significant.Immunohistochemical results showed that guanine had a therapeutic effect on hepatic fibrosis,and the effect of high dose and low dose was significant.Conclusion : 1.The related biochemical indexes of guanine after gavage were significantly lower than that of saline group,which indicated that guanine could protect the liver and improve the liver function of liver fibrosis induced by carbon tetrachloride.2.High-dose birds Purine to improve carbon tetrachloride-induced liver fibrosis in rats was significantly higher than the low dose group.
