Objective This experiment is to detect the expression of TNF-α and NF-kB in ratsthough TNF-α-mediated NF-kB signaling pathway, at the same time, to find theintervention and effect of rhodiola on cytokine expression in order to explore itsmechanism.Methods By random grouping method70healthy age-matched male SD rats weredivided into normal control group (C), model group(M), high-dose treatment group(G), middle-dose treatment group (Z), and low-dose treatment group (D).Except C,the rats of the four groups were subcutaneously injected of olive oil10%CCL4(5mL/kg),2times/week, total12W to establish liver fibrosis. Copy the model ofliver fibrosis, dosage and method are the same. Normal control group with salineinstead of10%CCL4.After twelve weeks, the rats of C and M were anesthetized andremoved liver tissue. From the9th week, the G, Z, and D groups rats were givenrespectively by Rhodiola rosea decoction. The dosage is3125mg/kg/day,312.5mg/kg/day,31.25mg/kg/day for12weeks. The rats of group G, Z and D wereremoved the fresh liver tissue specimens quickly and stored for later use at the end ofthe20th week The histopathological changes of liver tissues were observedby HE.The expression of statutory amount detection liver tissues TNF-αand NF-κB mRNA were detected by real-time quantitative PCR (Real-time-PCR).Results1.HE staining of liver biopsies showed: The liver tissue of M were damagedseriously compared with C, fatty degeneration of liver cells, necrosis and evenapoptosis; Fibrous tissue gaps became widen, leaflets structural were damaged,infiltration of inflammatory cells and false lobules were formatted. But, the livertissues morphology of G, Z and D group were improved with different degrees, inwhich the Z was obviously.2.RT-PCR results showed: The expression of TNF-αmRNA were C (0.06±0.03)、M(0.15±0.01)、G(0.12±0.01)、Z(0.07±0.02)、D(0.11±0.01),and NF-κ B mRNAwere C(0.06±0.03)、M(0.16±0.02)、G(0.15±0.02)、Z(0.07±0.03)、D(0.13±0.02). Compared with the C, The mRNA expressions of TNF-αand NF-KB in the M were significantly up-regulated (P<0.01); Comparedwith the M, The mRNA expressions of TNF-αand NF-KB in the G, Z, and D werereduced in varying degrees; the most significant was the Z (P<0.01);Conclusions1. The mRNA expression of TNF-αand NF-KB has changed significantly in liverfibrosis model, which may be associated with liver fibrosis and its progression.2. The effct of rhodiola on hepatic fibrosis in rats is significant in middle-dosetreatment group, which may be by inhibiting TNF-α-mediated NF-κB signalingpathway. Rhodiola may plays an important role in anti-fibrosis.
