Objective:Explore the association between the expression levels of membrane receptor HSPG2, ITGA6, ITGB1, CD151and the specificity of HPV types infecting cervical epithelial cells, aiming at providing a new path to clarify the mechanism of regional distribution difference of HPV types.Methods:1. qRT-PCR were used to detect the mRNAs expressions of HSPG2, ITGA6, ITGB1, and CD151in four cervical cancer-derived cell lines(SiHa, CaSki, Hela and C-33a). Western Blot were used to detect the proteins expressions of the membrane receptors.2. In152cases of HPV16,18,52,58infected and42cases of non-HPV infected cervical epithelial tissue, qRT-PCR were used to detect the mRNAs expressions of four kinds of membrane receptors above. Immunohistochemistry were used in further observation of the proteins expressions of ITGA6in401cases of cervical epithelial tissues which were under the infection of HPV16,18,52,58, and in33cases of cervical epithelial tissue without HPV infection.Results:1. The mRNAs and proteins of HSPG2, ITGA6, ITGB1and CD151were expressed in both SiHa, CaSki, Hela and C33a cell lines. But the expressions of HSPG2, ITGA6and CD151in C33a cell line were significantly lower than in the HPV-positive cell line and were highest in Hela cell line. ITGB1expressions were not significantly different between the four cell lines.2. In single HPV infected cervical tissues, the mRNAs expression levels of HSPG2, ITGB1, CD151is significantly higher than in HPV-negative tissues. The mRNAs expression levels of ITGA6was higher only in HPV-16,18, and52infected tissues, not in HPV-58infected tissues, compared with HPV-negative tissues.3. The ITGA6proteins were mainly expressed in the cell membrane and cytoplasm of basal layer of cervical squamous epitheliums. ITGA6proteins were significantly higher expressed in HPV-16,18and52infected tissues than HPV-negative tissues, but not in HPV-58infected tissues. These phenomenon could also be observed in morphological normal level, but not typical in higher levels.Conclusion:1. There were specificity of the types of infected HPVs in the expression of certain membrane receptors which play roles in HPV infection in the cervical epithelial cells.2. There are different forms of the secondary receptors expressions between HPV-16,18,52and HPV-58infected cervical tissues, suggesting the important roles of secondary receptors in the selection of certain HPV infection.3. The differences in the expression of membrane receptors occurred in the time before morphological changes were taken, suggesting the regulations by the receptors were occurred in the early stages of the virus infection.
