Screening Of Acupoint "Jieyufang" And Its Effect On HPA Axis Of Depression

Objective: In this study,on the basis of a large number of mining and summarizingthe literature, closely combined with clinical practice, the selected using high frequencyBaihui and Shenmen and Taichong three acupoint,analyze its influence on behavior of rat(Weight, Sugar water preference,Open-field Test),screening out the best formula acupointsin acupuncture treatment of depression,provide scientific laboratory basis for clinicalapplication. At the same time,using the best acupoints selected formula to study the effectof HPA axis of depression model rats on serum adrenocorticotropic hormone (ACTH) andcortisol (CORT) level,to explore the best formula acupoints mechanism for the treatmentof depression.Methods: First by Open-field to eliminate number of horizontal movement andvertical movement of less than30times or more than120times in the rat.Experiment one:Use90clean level SD male rats as the research object,randomly divide into: blankgroup(n=10),model group (n=10),acupuncture intervention group divide into9group,namely C～I group(n=10).9acupuncture intervention group:C group(Baihui group),Dgroup(Shenmen group),E group(Taichong group),F group(Baihui and Shenmen group),Ggroup(Shenmen and Taichong group),H group(Baihui and Taichong group),Igroup(Baihui and Shenmen and Taichong group).Model group and acupunctureintervention group rats adopt the combination of both long-term chronic unexpectedstimulation and solitary feeding,the rats have received21days of various stimulation.at thesame time,acupuncture intervention group before1hour in stress stimulation were givenacupuncture treatment.After the treatment,the rat behavior score were measured,screeningout the best formula acupoints in acupuncture treatment of depression andnamed“Jieyufang”.Experiment two: In experiment one, we have selected the best formulaacupoints,called the “Jieyufang”. On this basis,30clean level SD male rats as the researchobject were randomly divided into blank contrast group(n=10),model group(n=10), the“Jieyufang” group(n=10).modeling with experiment one,“Jieyufang” group before1hourin stress stimulation were given acupuncture treatment.Blank control group normalfeeding,Although model group by fetching, fixed, but do not do acupuncture,model groupwill be dealed same as the “Jieyufang” group but without electrical acupuncture treat,afterthe end of the treatment make rats anesthesia and draw blood4ml.Detect content of ACTH and CORT in rats,serum in each group by enzyme-linked immunosorbent assay(ELISA).Results:1.Behavior score results show:before the trial group rats,weight change, sugar waterconsumption,open-field Test results showed that no significant difference (p>0.05).(1)Weight change show:compared with the blank group,the7th day,the14thday,the21th day of the experiment,the weight of model group rats were decreasedobviously(p<0.05,p<0.01);compared with the model group,the7th day of theexperiment,the weight of Baihui group,Shenmen group,Taichong group,Baihui andTaichong group,Three point group rats were significantly increased(p<0.05), the weight ofBaihui and Shenmen group,Shenmen and Taichong group rats are also higher than modelgroup, but the results was not significant (p>0.05).the14th day of the experiment,theweight of each acupuncture intervention groups rats were significantlyincreased(p<0.05,p<0.01),the21th day of the experiment,the weight of each acupunctureintervention groups rats were increased obviously compared with the the14th day,amongthem,the weight of Three point group rats was significantly increased compared with therest of the acupuncture intervention groups(p<0.05).(2)The end of the experiment,the change of sugar water consumption:compared withthe blank group,the sugar water consumption of model group rats were decreasedobviously(p<0.01);compared with the model group,the sugar water consumption of Threepoint group rats was significantly increased(p<0.05),the sugar water consumption of therest of the acupuncture intervention groups are higher than model group,but the results wasnot significant (p>0.05).(3) The end of the experiment,open-field Test show:compared with the blankgroup,the open-field Test score of model group rats were decreasedobviously(p<0.01,p<0.05); compared with the model group,the open-field Test score ofeach acupuncture intervention groups rats were significantlyincreased(p<0.05,p<0.01),among them,the open-field Test score of Three point group ratswas significantly increased compared with the rest of the acupuncture interventiongroups(p<0.05).2. For the influence mechanism of EA acupoints“Jieyufang”to HPA axis of depression model rats study results show:compared with the blank group,the ACTH and CORTcontent of model group was significantly increased(p<0.01);compared with the modelgroup,the ACTH and CORT content of acupoints“Jieyufang”group decreasedobviously(p<0.01).Conclusion:1.Screening out the best formula acupoints in acupuncture treatment of depressionis:Baihui and Shenmen and Taichong.2.EA acupoints“Jieyufang”can obviously adijust the content of ACTH and CORT inrats serum,so therapeutic mechanism of acupoints“Jieyufang”maybe by way of adjust thefunction of HPA axis.
