| Objective:This study by electric acupuncture acupoints “Jieyu”formula depression rats with chronic unpredictable mild stress,compared with model group and acupuncture group rats behavioral changes,such as the consumption of sugar water changes,the changes of weight,curious about the outside world degree of change,the change of space exploration ability in the water maze and serum IL-1 beta,IL-6 and TSH level expression from several aspects,such as the change of the explanation from the macro acupoints "Jieyu" changes in behavior of depression model rats,from microscopic research on acupoints "Jieyu" for depression rats immune system and endocrine system,the influence of explore curative acupoints "Jieyu" the mechanism of the treatment of depression,to promote clinical acupoints "Jieyu" treatment of depression to provide more reliable laboratory basis.Methods:Selected five minutes through open-field test times the sum of the number of horizontal movement and vertical movement in 30 rats 30-120 times,the rest of the strip,then 30 SD rats were randomly divided into 3 groups,each group of 10.Group A: blank group;Group B: model group;Group C: acupuncture group.Group A as A blank group,regular feeding,without any treatment;Group B and group C and begin to accept every day around the timing of different stress: cold water swimming(4 ?,5 min)and banning water(24 h),fasting day and night(24 h),reverse(24 h),damp bedding(24 h),binding(3h),clip tail(3 min).The above stress stimuli were randomly selected each day,and each stress pattern was used on average four times.Group A does not offer stress stimulation and acupuncture treatment,group B took chronic unpredictable mild stress and not given acupuncture treatment,group C immediately take acupuncture treatment after stress.disinfect acupoints and the zhongyantaihe 0.25mm×25mm acupuncture needle was selected for acupuncture,use SDZ-II electric acupuncture apparatus to strengthen stimulation.Using electric acupuncture apparatus not excessive stimulation when the rats in the struggle,bray,in general with the point of skin around vibratiuncle is appropriate,the current should be kept below 2mA,for 30 min,1 times / day,acupuncture treatment of28 d..At the end of the experiment,the results of laboratory data comparison of rats were obtained by the behavior of rats.Serum inflammatory factors and thyroid hormone levels in rats were detected by enzyme-linked immunosorbent assay(ELISA).Results:1.we can see that there was no significant difference in weight,sugar water consumption and open field test in the group A,group B and group C before the experiment(P >0.05).(1)weight change after 28 days: compared with group A(blank group),the weight of rats in group B(model group)decreased significantly(P<0.01).Compared with group B(model group),the weight of rats in group C(acupuncture group)increased significantly(P<0.01).(2)after 28 days,the sugar water test situation: compared with group A,the consumption of sugar water in group B was significantly reduced(P<0.01).Compared with group B,the sugar water preference in group C was significantly increased(P<0.01).(3)after 28 days,the open field test situation: compared with group A,the horizontal and vertical motion scores of group B significantly decreased,and the results were significantly different(P < 0.01).Compared with group B,the number of horizontal and vertical movements in group C increased significantly(P < 0.01).(4)water maze test in the late experiment: compared with group A,the spatial exploration ability and learning and memory ability of group B were both decreased(P < 0.01).Compared with group B,rats in group C significantly improved their learning and memory ability and spatial exploration ability(P < 0.01)2.The effect of acupuncture on the serum of rats: Compared with group A,the serum IL-1?,IL-6 and TSH levels in group B changed significantly(P < 0.05).Group C compared with groupB,serum IL-1? and IL-6 levels cut down and TSH levels increase,there is significant difference(P < 0.01),compared with the group A,there was no significant difference in group C(P > 0.05)Conclusion:1.The CUMS model can successfully simulate depressive symptoms similar to humans in animals,with symptoms such as slow weight gain,loss of pleasure,decreased mobility,decreased curiosity,and depressive symptoms that show a dull response to spatial memory.CUMS model rats showed more obvious immune stress response and neuroendocrine metabolic disorders,mainly in the serum levels of pro-inflammatory cytokines IL-1?,IL-6 levels and TSH levels decreased.Results:3,Electroacupuncture points "Qiyu prescription" can significantly improve the depressive symptoms in rats.4.Electro-acupuncture points "Qiyu prescription" can reduce the serum IL-1?,IL-6content,TSH content increased.5.Electro-acupuncture point "Qiyu prescription" can improve learning and memory ability and space exploration ability of depressed rats.To sum up,the mechanism of treating depression by electric acupuncture is related to regulating the secretion of inflammatory factors in serum and regulating hypothalamic-p ituitary-thyroid axis function. |
