Purposes:This subject adopts a high-fat diet copy dampness heat stasis of nonalcoholicfatty liver disease in rats model, by using HE staining method to detect the experimentbuilding success or failure, the application of ELISA (enzyme-linked immune sorbent assay)detection of liver lipid soluble granules on rat liver tissue pathology and rat serum TNF alpha(tumor necrosis factor alpha) expression. TNF-α and nonalcoholic fatty liver disease, therelationship between argument liver lipid soluble granules on the mechanism of action ofintervention of nonalcoholic fatty liver disease.Methods:60, clean level Wistar rats to give ordinary feed, with7days were randomlydivided into2groups of laboratory environment, high fat group and51, common set of nineonly. High fat group was given high fat feed (feed composition:10%lard,2%cholesterol,88%feed) feeding, ordinary group gives ordinary feed. After12weeks, in the fatty group andordinary group to pick one with6rats, immediately after the death in liver tissue for HEstaining, observation of building success or not. Remaining group rats (9) of your normalfeed. The remaining high lipid group rats were randomly divided into5groups: model group, control group, high, medium and low dose group,9rats in each group. Including Chinesemedicine high, medium and low dose group and control group in the daily continue to highfat feed given different drugs at the same time to fill the stomach; Normal group and modelgroup was given saline lavage (LML/100g). Control group given to the concentration of0.14g/kg of polyene phosphatidylcholine capsule aqueous solution to fill the stomach; Groupwere given different doses Chinese materia medica liver lipid soluble particles aqueouslavage (concentration respectively:3g/kg,2.1g/kg,0.9g/kg). These six groups of ratsweight, daily activities, eating, and so on and so forth. After4weeks continuous lavage, livertissue line HE staining, observe the liver pathology morphology change; Take artery(abdominal aorta) using the enzyme linked immunosorbent assay serum TNF-α expression.Results:1. The liver lipid soluble granules on the influence of NAFLD rat liver indexHigh dose group of liver index was significantly lower than control group (P <0.05); Dosegroup in liver index compared with control group, the difference (P>0.05); Low dose groupof liver index was significantly higher than that of high, middle dose and the control group (P<0.05). 2. The liver lipid soluble granules on the influence of NAFLD rat liver pathologicalmorphologyDegree of model group rats liver tissue fatty change compared with the normal group hadsignificant difference (p <0.01); Application of drug lavage rats (control group, high, mediumand low dose group) of the pathological change of liver tissue has different degrees ofimprovement, hepatocyte fatty change number of lipid droplets in cytoplasm and rate reducedto some extent, is better than that of model group significantly (p <0.05); Traditional Chinesemedicine (TCM) rat liver tissue pathology to improve high dose group was obviously betterthan that of control group (p <0.05); Traditional Chinese medicine (TCM) in the dose grouprat liver steatosis degree compared with control group, no significant difference (p>0.05);Liver fat soluble high and low dose group of Chinese medicine, middle dose group and thecontrol group, compared with significant difference (p <0.05).3. The liver lipid soluble granules on NAFLD rat serum TNF-α research on the effects ofexpressionThe model group rats serum TNF-α expression is significantly higher than normal group (P<0.01), statistically significant; The expression of the model group rats serum TNF-α issignificantly higher than the drug intervention group (P <0.01), statistically significant; Thehigh dose group of rats serum TNF-α expression significantly lower than control group (P <0.05), statistically significant; Dose group of the expression of rat serum TNF-α comparedwith control group (P>0.05), there was no significant statistical significance; Low dosegroup of the expression of rat serum TNF-α compared with high, middle dose group and thecontrol group (P <0.05), with statistical significance.Conclusion:1NAFLD rat feeding hepatic lipid soluble granules can improve the general condition of theoriginal has been sick, significantly reduce liver lipid deposition, improve the degree ofhepatic steatosis.2hepatic lipid soluble particles significantly decreased the serum TNF-α expression.3hepatic lipid soluble granules by reducing lipid deposition in the liver, improve the degreeof hepatic steatosis, inhibit expression of serum TNF-α, regulate lipid metabolism, promotethe decomposition of fatty acid oxidation, reduce liver steatosis, thus play a role of treatment of nonalcoholic fatty liver disease, which may be liver lipid soluble granules one of themechanism of prevention and treatment of nonalcoholic fatty liver disease.
