OBJECTIVE:To establish a model of NAFLD with diabetes induced by high-fat diet combined with streptozotocin,and to observe the effect of Ganzhirong granules on serum biochemistry and liver morphology of Wistar rats.The effects of fatty liver and glucose and lipid metabolism in diabetic rats.METHODS:The purchased rats were housed in a laboratory and given ordinary pellets.During the experiment,the animals were given free access to water.Rats were randomly divided into normal group and experimental group after 7 days of adaptation to the environment.The normal group was fed with normal diet.The experimental group was fed with high-fat diet.After 12 weeks,the rats were fed with high-fat diet.The experimental group was intraperitoneally injected with 1%STZ 30 mg·kg-1.The normal group was intraperitoneally injected with equal volume of citric acid.-Sodium citrate buffer.The successful model rats were divided into model group,high-,medium-,and low-dose group of Ganzhirong granules and metformin hydrochloride group,with 10 rats in each group.The normal group and the model group were given normal saline(1ml/100g)daily;the high,medium and low dose groups of Ganzhirong granules were given different concentrations(100%,70%,30%)of Ganzhirong granules aqueous solution.Gastric administration(3g/kg,2g/kg,1g/kg).The control group was given metformin hydrochloride,and the mixture was stirred until completely dissolved,and then diluted with distilled water at a dose of 200mg/(kg·d)-1.After 12 weeks of the experiment,blood was collected from the abdominal aorta and taken.The liver pathological tissues of rats were observed and serum TG,TC,ALT,AST,GGT,FBG,FINS and IR were detected.Results:1 The liver index of the high-dose group was significantly lower than that of the metformin group(P<0.05).2 HE staining was observed under light microscope:the model group had the highest degree of hepatic steatosis,and the normal group had the lowest degree of hepatic steatosis.The degree of hepatic steatosis in the high-dose group was significantly lower than that in the control group(P<0.05).3 The serum levels of TG,TC,ALT,AST,GGT,FBG,FINS and IR in the high dose group of Ganzhirong granules were significantly lower than those in the metformin control group(P<0.05).Conclusion:1 Ganzhirong granules can improve the general state and liver index of NAFLD combined with T2DM rats,reduce lipid deposition in hepatocytes and reduce steatosis.2 Ganzhirong granules have the effect of lowering serum transaminase(ALT,AST,GGT),improving liver function,protecting liver cells,to the treatment of NAFLD combined with T2DM.3 Ganzhirong granules have the effect of lowering serum TG,TC content,improving lipid metabolism disorder,avoiding lipid deposition in liver cells,and protecting the liver,thereby treating the combination of NAFLD and T2DM.4 Ganzhirong granuleshave the effect of increasing insulin sensitivity,regulating the level of glycolipid metabolism,can effectively treat NAFLD combined with T2DM.
