Objects:To investigate the therapeutic effect and mechanism of Huzhang granules on rats with nonalcoholic fatty liver disease (NAFLD) induced by high-fat feeding.Methods:Fifty male SD rats were randomly divided into two groups:control group (A), ten rats were fed with normal diet; model group, forty rats were fed with high fat diet. After four weeks, the rats in model group were fed with high fat diet consecutively and randomly divided into4subgroups:model group (B) and three therapeutic groups (C1, C2and C3), which were administered with normal saline, Huzhang granules11.275g/kg(C1),22.55g/kg(C2) and45.1g/kg(C3) daily respectively. All rats were sacrificed after four weeks. Serum, liver and adipose tissue were preserved for biochemical assay and histological examination. Expression of PPARy coactivator-1, PPARa and Liver X receptor-a mRNA were detected by real-time PCR.Results:High dose of Huzhang granules decreased serum ALT and lipid levels. This medication up-regulated the expression of PPARĪ³ coactivator-1and PPARa mRNA in a dose-dependent manner, and the expression of Liver X receptor-a mRNA was dose-dependent down-regulated as well.Conclusion:Huzhang granules have effect on the treatment of NAFLD. It may regulate the deposit of fatty acid in liver and adipose tissues by regulating the expression of gens involved in the metabolism of fatty acid, resulting in lowering serum lipid and improving liver function, and preventing NAFLD.
