Objective: To observe the effects of the lipid-soluble extracts ofMortierella isabellina on rats suffering from the experimental nonalcoholicfatty live diseases induced by diet rich in lipid. Methods: 32 Wistar rats were divided into four groups: normalgroup, model group, lipid-soluble extracts of Mortierella isabellinatreatment group and Hepatopoeitin(HPN) group at random.The normalgroup fed on normal diet ,while the model one ,treatment one and theHPN one were given the diet abundant in triglycerides andcholesterol.At the same time ,the treatment and HPN group wereadministered orally the lipid-soluble extracts of Mortierella isabellina andHPN respectively,the normal and model group were administered orally thenormal saline. After 12 weeks ,The changes of TG and TC in both serumand liver tissue,CHE and Aminotransferase in serum,SOD and MDA inliver and liver-to-body weight ratios were measured.on the other hand ,thegrade of hepatic steatosis and Immunohistochemical results of TNF-а andCYP2E1 were also evaluated. Results: 1.TG and TC in serum and liver, CHE,ALT and AST in serum andliver- to- body weight ratios decreasedby18.4%,19.4%,22.9%,27.1%,23.5%,27.7%,37.8%and18.6% ,respectively,SOD increased by 34.2% while MDA reduced by 22.1% in liver,theAIOD of TNF-α and CYP2E1 decreased by 52.2% and 56.3%immunohistochemically ,when compare the treatment group with the modelone. (P﹤0.05 or ﹤0.01 between all two groups ) 2. In the case of comparing the treatment group with the normal TGand TC in serum and liver, CHE,ALT and AST in serum and liver tobody weight ratios increased by 29.9%,28.8%,16.1%,46.9%,54.8% ,63.8%,40.3% and 23% respectively,SOD was lower by15.5% while MDAhigher by 22.4% in liver, the AIOD of TNF-αand CYP2E1 increased by267% and 50% immunohistochemically. (P﹤0.05 or ﹤0.01 among alldata cited of two groups ) 3.Turn to compare the model group with nomal one , TG and TC inserum and liver, CHE,ALT and AST in serum and liver- to- body weightratios raised 59.7% , 55%,38.2%,136.4%,101%, 125%,83% and57.4% ,respectively,SOD lessened by 37.1% while MDA hightened by57.1% in liver. the AIOD of TNF-αand CYP2E1 increased by 667% and243% immunohistochemically.All difference between two groups citedabove are markly in statistics .The degree of hepatic steatosis in liver oftreatment group was less serious than model one,but more than normal one.Hepatopoeitin shew no effect on lessening the lipid in serum orliver ,CHE in serum or the hepatic steatosis,while lowering the content ofaminotransferase in serum and MDA in liver ,and AIOD of TNF-α andCYP2E1 immunohistochemically,elevating the content of SOD in liver. Conclusions: The lipid-soluble extracts of Mortierella isabellina canprotect the rat fed on diet abundant in lipid from NAFLD as well as preventthe excess of lipid from depositing in serum or liver.Moreover,it exihbitsthe effect on weakening the oxidative stress or lipid perioxidation .WhlieHPN only showed the effect on weakening the oxidative stress or lipidperioxidation and no effect on decreasing the the excess of lipid in blood orthe liver.
