The animal skin, an important protective barrier against the mechanical injury and microbial attack, is the body’s largest organ. The healing of the mechanical injury of the skin is a key to the question in the clinical study, and it is divided into three stages: the inflammatory response stage, the development stage of granulation tissue, the skin regenerative stage. The quantity and quality of granulation tissue will influence the healing degree of the mechanical injury of the skin and the outcome. TGFβ1 and VEGF, as important cytokines in the stage of granulation tissue eproductive, play a key role in all the process of skin wound healing. In this case, the healing degree could be observed by the change of protein and m RNA expression levels. Although it is a century to study the mechanism of the medicine with the mechanical injury of the skin, yet it is not found a effective medicine to cure the mechanical injury of the rats’ skin.The Chinese Traditional Medicine is redeveloped in recent years with it’s abundant resources.The mirabilite, documented in “Bie Lu”, have a therapeutic of the diarrhea, the constipation, et al. Modern biology considers that the mirabilite could promoted angiogenesis and tissue regeneration after the injury of skin. More and more researchers invest their attention to this medicine in the mechanical injury of skin healing.Here we used the mechanical injury of the rats’ skin models in this study to study alterations in gene and protein expression of rat treatment via the external application with mirabilite. The genes expression of cytokine VEGF and TGFβ1 in the mechanical skin injury rat in vivo and inoculated EA.hy926 cell and L929 cell in vitro were analyzed by q RT-PCR and enzyme-linked immunosorbent assay(ELISA) at different 2 day,5 day and 9 day. In addition, the effect of mirabilite were evaluated by the wound area measurement. Our results show a marked effect of the curative effect of medication on the m RNA expression of cytokine(VEGF and TGFβ1) of the mechanical skin injury rat. In addition, the highest healing percent of the skin injury rat was 79.01%. Moreover, in vitro, the mirabilite(10-100 μg/ml) in EA.hy926 and929 cells, induce the secretion of VEGF and TGFβ1. Together, this results indicated that the expression level of the VEGF and the TGFβ1 genes and protein that the healing to the mirabilite in vivo and vitro of rat.
