Objective: A rat model of wound healing was established,and wound with mirabilite external use.Wound healing rate and histological changes were observed at different time points in the healing process.In addition,the infiltration of macrophages and the expression of VEGF and TGF-? 1 in the wound tissue were observed.Objective to investigate the effect of Mirabilite external use on wound healing and analyze its mechanism from cellular molecular perspective.Methods: A total of 60 male SD rats were randomly divided into model group,control group,mirabilite group,each group of 20 SD rats were established with diameter about 2cm in the rat dorsal side of a circular full-thickness skin wound model.On the first day after injury,the wounds of model group,control group,mirabilite group were given without drug dressing,1%DMSO dressing,mirabilite solution dressing intervention respectively.Wound healing was observed at 3,5,7,10,14 days after injury,meanwhile,the wound surface area was measured by membrane tracing,and the rate of wound healing was calculated.4 rats were randomly executed at each time point.The wound tissues were stained with HE to observe the histological changes of wound healing and to calculate the cross sectional area of the new capillaries,in addition,Masson staining was used to observe the proliferation of collagen fibers in the three groups.Immunohistochemical staining was used to label macrophages,VEGF and TGF-? 1.At high magnification,the infiltration of macrophages and the expression of VEGF and TGF-? 1were compared in the three groups at each time point by cell count and mean light density.Results:It was found that the filling and contraction of the wounds in the mirabilite group were faster than that of the model group and control group with view of the naked eye.The rate of wound healing in the three groups was compared.the difference in average wound healing rate between the model group and control group was not statistically significant at the five time points(P > 0.05).3 days after injury,the average wound healing rate of mirabilite group was higher than that of control group,but the difference was not statistically significant(t=1.45,P=0.154).At 5 and 7days after injury,the average wound healing rate of mirabilite group was higher than the control group,the difference was statistically significant(t=2.11,P=0.043;t=2.11,P=0.046).At 10,14 days,the average wound healing rate of mirabilite group is higher than that of control group,but the difference was not statistically significant(t=1.40,P=0.183,t=0.79,P=0.461).The wound tissue HE staining showed that the mirabilite group,model group and control group showed no difference in the histological changes at 3 days after injury.At 5,7 days after injury,compared with the model group and control group,the mirabilite group showed that the new vascular tissue is more abundant,the number of cells more compact,and the density of tissue is higher.At 10,14 days after injury,The wound tissue in the mirabilite group was more compact than that in the model group and control group.Comparing the cross-sectional area of the new capillaries,the difference between the model group and control group was not statistically significant at each time point(P > 0.05).At 5,7 days after injury,the mean cross-sectional area of new capillaries in mirabilite group was larger than others,the difference was statistically significant(P < 0.05).At 3,10,14 days after injury,there was no significant difference in the cross-sectional area between the three groups.The results of Masson staining showed that the density and thickness of the collagen fibers in the mirabilite group were greater than the model group and the control group at 5,7 days after injury.The immunohistochemical results showed that there was no significant distinction in the number of macrophages between the model group and control group at each time point after injury(P > 0.05).The mean value of macrophage cells count in the mirabilite group was higher than the model group and control group at 3,5 and 7 days after injury.The difference was statistically significant(P < 0.05).Although the mean value of macrophage count in mirabilite group was larger than that in model group and control group at 10,14 days after injury,the difference was not statistically significant(P > 0.05).The expression of VEGF and TGF-? 1 in the three groups was higher at the 3,5 and 7 days after injury,but the expression decreased at the 10,14 days.The comparison results of optical density value show that there was no significant difference in the mean optical density of taht VEGF and TGF-? 1 expressed between the model group and control group during the wound healing process(P > 0.05).At 3,5,7 days after injury,the mean optical density of VEGF and TGF-? 1 expressed in the wounds of mirabilite group was obviously higher than model group and control group,the difference was statistically significant(P < 0.05).At 10,14 days after injury,although the mean optical density of VEGF and TGF-? 1 in mirabilite group was higher than that in model group and control group,there was no significant difference(P > 0.05).Conclusions:The external application of mirabilite can promote the Neovascularization,the proliferation of various cells and the proliferation of collagen fiber in the process of wound healing,so as to promote the filling and contraction of wounds and accelerate the healing of wounds.External use of mirabilite can increase the infiltration of macrophages and the expression of VEGF and TGF-? 1 in the wound tissue at the early stage of wound healing(inflammatory and proliferative stages).The regulation of macrophage infiltration in the wound may be the main mechanism for the external use of mirabilite to promote wound healing.
