Study Of Juan Bi Particles Interventing Rabbit Knee Joint Cartilage Cell Apoptosis Based On The Level Of Caspase-3

Objective: Through controling the balance of caspase-3 protein expression, discusses the influence of Juan bi particles to rabbit knee osteoarthritis cartilage chondrocytes cell apoptosis, to reveal a part of the mechanismthe of osteoarthritis(OA) treatment, and provides the theory basis for treatment of OA.Methods: using improved Hulth method to establish the rabbit model of knee OA, will the experimental rabbits were randomly divided into control group, model group, Juan bi granule group and the positive drug group. after 6 weeks drug intervention, make HE dyeing paraffin sections and observe the pathological morphological changes of cartilage tissue under the mirror, to detect cartilage caspase-3 expression level by immunohistochemical method, and detect serum NO level by ELISA,to detect chondrocytes cell apoptosis by TUNEL method.Results: 1. Juan bi particles can cut the caspase-3 protein expression of rabbit knee joint cartilage, compared with model group, positive control group, the positive cell rate is significantly lower than the model group and positive medicine control group(P < 0.05);2.Juan bi particles can decrease the cell apoptosis rate of rabbit knee joint cartilage, compared with model group, cartilage cell apoptosis rate of Juan bi particlesis group is lower than the model group(P < 0.05); 3.Juan bi particles can reduce the serum NO, IL- 1β levels of rabbit knee osteoarthritis model, compared with model group(P < 0.05).Conclusion: 1. Juan bi particles could modulate the caspase-3 protein expression, reduce the level of serum NO, IL- 1β, inhibit apoptosis of chondrocytes, and has a protective effect on knee joint cartilage.2. Inhibiting caspase-3 protein expression and reducing the cartilage cell apoptosis, is likely to be one of the mechanisms of Juan bi particles treating OA.
