Objective:To study the mechanism of Qingmaiyin and its decomposed formulas itsdecomposed formulas by interposing NF-κB signaling pathway on atherosclerosis obliteransrabbit.Method: Selected SPF grade male rabbits for the study, by high-fat diet, immune dysfunctiondamage and the right femoral artery ligation ASO established animal model,60rabbits wererandomly divided into Quanfang group, Ruanjian group, Huoxue group, Qingfa group,simvastatin statin group, prevention group, model group, control of model group, fake modelgroup and control group. The prevention group was intervention by Qingmai drink the wholeperiod, and at the time of modeling was completed, the rest of the group with normal diet, andintervention with water and corresponding drug executed after8weeks. To detect theexpression of TNF-αmRNA, I-κBmRNA and NF-κBmRNA in aortic endothelial tissue withReal-time quantitative PCR (QPCR). Using automatic biochemical analyzer to measure fourserum lipids. Measure the expression levels of serum CRP and ET-1with Elisa kit.Results: The expression of TNF-αmRNA does not have significant difference (P>0.05). Theexpression I-κBmRNA among the Quanfang group, Ruanjian group, Huoxue group weresignificant differences (P <0.01), but do not have difference with control group. Theexpression of Qingfa group, simvastatin group and prevention group were higher than thecontrol of model group and model group(P <0.01),and had significant difference with thecontrol group(P <0.05);the expression of Qingfa group, simvastatin group and prevention group were higher than Quanfang group, Ruanjian group and Huoxue group (P <0.01).In theexpression of NF-κB mRNA, the Ruanjian group, simvastatin group and Qingfa groups werelower than model group and the control of model group, while higher than the control group,and they were significant differences (P <0.01), the expression of Quanfang group andHuoxue group lower than the model group, while higher than the control group, and they weresignificant differences (P <0.01), there was no significant difference with the control group(P>0.05), there were no significant difference between Ruanjian group and simvastatin group(P>0.05), there were significantly different among Ruanjian group, Quanfang group, Huoxuegroup and Qing groups (P <0.01), Huoxue group and Quanfang group were not differencewith the model of low-fat diet control group (P>0.05), the expression of the interventiongroup lower than the model group, while higher than the control group, they were significantdifferences (P <0.01), there was difference with the control of model group (P <0.05), exceptfor Ruanjian group(P<0.05), there were no differences with other group(P>0.05).Theexpression of HDL, simvastatin group was the lowest, and has significantly difference withQuanfang group, Ruanjian group, Qingfa group and Huoxue group(p <0.05). The expressionof downregulated arteriosclerosis index, Quanfang group, Huoxue group and Qingfa groupwere more than simvastatin group (p <0.05). The expression of CRP,Qingfa group was thelowest. There were significant differences with the control of model group(p<0.01),simvastatin group followed with the control of model group also significantly different withthe control of model group (p <0.05), there was significantly difference between Ruanjiangroup and control group(p <0.05), the expression of Huoxue group and the control of modelgroup are also significant differences (p <0.05). Huoxue group is the lowest on the expressionof ET-1, and significantly difference with control group, model group and the control of modelgroup(p <0.05), followed by Qingfa group, there were significant difference with the modelgroup and the control of model group (p <0.05). Conclusion:1. the intervention of NF-κB signaling pathway on arteriosclerosisobliterans rabbit of Qingmaiyin and its component was not just rely on TNF-αviaNF-κB classical pathway to achieve, but it was still mainly by adjusting theexpression of I-κB to achieve.2. Viewed from all angles in the experiment, the effectof Qingfa group was the best in Chinese medicine Qingmaiyin and its decomposed formulas.
