Object:1、 Establishing the experimental rabbit models of hypertension andatherosclerosis of brain;2、Study the relationship between blood pressure variabilityand atherosclerosis of cerebral artery in rabbits.Method:Twenty-six New Zealand White Rabbits were randomly divided into threegroups:Group A: Drug intervention group (blood pressure increased after modelEstablished:n=11),Group B:placebo group (blood pressure increased after modelEstablished,n=6),Group C:sham-operated rabbits(high fat diet feeding,n=6),Group D:normal control group (common feeding,n=3);Measuring the bloodpressure of the rabbits once in every five days.As soon as the blood pressure raisingafter surgery,we started to use the antihypertensive drugs to intervene the pressureand feed high-fat diets at the same time.The rabbits were regarded as the hypertensionmodels after the blood pressure reaching the Standard of high blood pressure. wemonitored the blood pressure every five days and monitored the B ultrasound ofcarotid artery.After10weeks,we took the fresh left carotid arterys after anesthesia forPCR from the rabbits to detect the expression of CRP mRNA.After the above stepswe used the constant pressure perfusion to fix the right carotid artery and thoracicaortic artery for HE staining.Result:We get the results,the median of blood pressure SD value of group A wasdivided into two groups, group a is greater than the median arrays and group b lessthan the median.Group B(placebo) regard as group c,group C (control group) regardas group d,group D (normal group)regard as the group e.1、Compared each group with blood pressure variability(the blood pressure SD as theblood pressure variability).In the experiments,there are significant differencesbetween group a and group b,group a and c,group a and d,group a and e,there are differences in the DBP and MBP of group b and group d,group c and group e.2、Compared the results, of carotid artery IMT of the B ultrasound between the fivegroups in ten weeks, showed no significance. There was no significant differencebetween these groups in the zero-th week and the tenth week of carotid artery.3、Compared the relative expression of CRP mRNA of left carotid artery among thesegroups,It could be saw obviously differences in group a and group b, group b andgroup c, group b and group d,group b and group e.Group a and group b, group b andgroup c, group b and group d, group b and group e had no obviously differences inthem.4、The pathological examination mainly compared the aorta I/M value in theexperiments,there were differences in group a and group c,Group c and groupd.There were no statistical difference among other groups.There were no significantdifferences in the I/M value of carotid artery among the five groups.Conclusion:1、We can produce good experimental rabbit models of hypertension andatherosclerosis of brain;2、The carotid artery IMT of the B ultrasound can be used asthe indicator of the research of the relation between bloodpressure variability and atherosclerosis;3、This rabbit model can be used to study therelationship between the blood pressure variability and atherosclerosis.
