Objective: The basic pathogenesis of CFS, to establish a rat model of chronic fatigue,observed Guilu Yishen Granule on rats with chronic fatigue in general and behavioralindicators (vertical movement, cross grid exercise, exhaustive swimming time)influence, determination of chronic fatigue in skeletal muscles of rats, to observe theeffect of Yishen granules of Guilu the content of PGC-1α in the skeletal muscle ofrats with chronic fatigue, further explore its mechanism, provide the basis ofexperiment and support of theory to the clinical application of Guilu Yishen granule inthe treatment of CFS.Methods: male SPF SD rats:40, weight:200±20grams. After1week adaptivefeeding, divided into model group and non group, model group with24rats, not createmodule16. After the success of modeling, not molding were randomly divided intotwo groups, including normal group and normal control group; model were randomlydivided into3groups, respectively, model group, Congrong Yishen granule group,Guilu Yishen Granule granule group, with8rats in each group. Chronic restraint,provoked and exhaustive swimming method to construct the model of rats withchronic fatigue. The normal group, the normal control group was given ordinary feed;model group, Congrong Yishen granule group and Guilu Yishen granule groupaccording to the method of modeling,14D. After modeling, the rats weight growth isslow or reduce, drinking, eating less, loose stools, rat hair dull dull, confirmed that therat model of chronic fatigue model. After making the mould by gavage, the normalgroup and the model group according to the10ml/(kg d) weight given normal saline,normal control group and Guilu Yishen granule group according to1250mg/(kg d)give Guilu Yishen granule suspension gavage, Congrong Yishen granule groupaccording to the417mg/(kg D) given particle Cong Rong Yishen suspension gavage,a total of1times a day, continuous intragastric administration of14days. Beforemodeling, after modeling and after gavage respectively measured every weight,vertical movement times and cross lattice exercise frequency and time of exhaustive swimming.Results:(1) after modeling, make weight model rats to reduce or slow growth, waterconsumption, decrease of eating, fur dull dull, reduced activity. Don’t make weightmodule rat normal growth, fur bright color, normal activities. Compared with thenormal group, control group, Congrong Yishen granule Guilu Yishen granulestreatment group, model rats decreased food intake, rat hair dark color, weight loss,vertical motion, reducing the number of cross grid exercise, exhaustive swimmingtime shorter, there were significant differences (P<0.05). After intragastricadministration, compared with the normal group, normal control group and GuiluYishen granule weight group, exhaustive swimming time, vertical motion, cross gridmovement times without significant difference (P>0.05). Compared with the modelgroup, the weight of Congrong Yishen granule group, Guilu Yishen granule group rats,a significant increase in the number of cross grid exercise, exhaustive swimming timewas significantly prolonged, there was significant difference (P<0.05). Guilu Yishengranule group and Congrong Yishen granule group rats were compared, weight andvertical motion, cross grid motion were no significant difference (P>0.05), exhaustiveswimming time was significantly different (P<0.05).(2) Compared with the normal group: normal control group, model group, thecontent of PGC-1α and Congrong Yishen granule group in skeletal muscles of ratshad significant difference (P<0.05); there was no significant difference in the contentof PGC-1α Guilu Yishen granule group in rat skeletal muscle (P>0.05). Comparedwith the model group: the increasing content of PGC-1α Guilu Yishen granule groupand Congrong Yishen granule group in skeletal muscles of rats, there were significantdifferences (P<0.05); compared with Congrong Yishen granule group: the increasingcontent of PGC-1α Guilu Yishen granule group in rat skeletal muscle, the difference(P<0.05).Conclusion:(1) The chronic restraint, provoked and exhaustive swimming and othercomplex factors, establishment of a rat model of chronic fatigue and chronic fatiguestate, can simulate human, and strong operability, feasibility.(2) Guilu Yishen granule can improve the vertical movement of rats with chronicfatigue and the number of cross grid movement times of rats with chronic fatigue,prolong the exhaustive swimming time, improve the physical and mental fatigue andchronic fatigue rats, and provide experimental basis for clinical application of GuiluYishen granule in the treatment of CFS. (3)The experimental results show that, PGC-1α in skeletal muscle of rats with chronicfatigue decreased significantly, PGC-1α in Guilu Yishen granule group wassignificant higher in skeletal muscle. Clarifies the effect of anti-fatigue benefits GuiluYishen granule.
