Objective:By preparing the rat model with chronic fatigue,we focuse on the basic of pathogenesis of CFS and observe Shengyang Yiwei Particles on chronic fatigue rats general situation and behavioural indicators(exhaustion swimming time,standing movement and across the grid times);observe the effect of Shengyang Yiwei Particles on the contents of SDH and Cyt-c of chronic fatigue rat skeletal muscle mitochondria,as to probe the anti-fatigue action mechanism of Shengyang Yiwei Particles,and provide the experimental and theoretical evidence for the treatment of CFS with Shengyang Yiwei Particles in clinical application.Method:(1)Grouping,Modeling:After adaptive feeding 3 days,we randomly divided 60 SPF SD male rats into:normal group,model group,Nuodikang Capsule group,Shengyang Yiwei Particles low dose group,Shengyang Yiwei Particles middle dose group,Shengyang Yiwei Particles high dose group,each group had 10 rats.The normal group without modeling and the other 5 groups carried modeling,time for 14 days.We adopted forced chronic bondage,exhaustion swimming and clip tail composite method to construct the model of rats with chronic fatigue.(2)Method of administration and dose: Ending the modeling,the normal group and the model group according to the 10ml/(kg · d)given normal saline,the Nuodikang Capsule group according to 0.182g/(kg · d)given Nuodikang Capsule suspension gavage,the Shengyang Yiwei Particles low dose group according to 1.625g/(kg · d)given Shengyang Yiwei Particles suspension gavage,the Shengyang Yiwei Particles middle dose group according to 3.25g/(kg · d)given Shengyang Yiwei Particles suspension gavage,the Shengyang Yiwei Particles high dose group according to 6.5g/(kg · d)give Shengyang Yiwei Particles suspension gavage.Rats in each group were administered 1 times a day,for 14 consecutive days.We weight each rat every 3other day so as to adjust administered dose.(3)Extract tissue: After successful local anesthesia in rats,we took the piece of left hind limb skeletal muscle(1g)of rats in each group,and rinse in normal saline to prepare skeletal muscle tissue homogenate and mitochondria suspension,then packing label keeped in the-20? freezer.(4)The detecting of experimental indicators:We measured the rat body weight,exhaustion swimming time,across the grid and standing movement times in the day before modeling,after modeling and after the last administration.We evaluated the contents of SDH and Cyt-c of rat skeletal muscle mitochondria by ELISA method.We used Sp SS18.0 statistical software to process the collected data.Results:(1)General Situation: before modeling,there are no differences in weight,eating and drinking,movement,fur,and feces between each group of rats.After modeling,with time passed by,except the normal group,the other group rats lost weight little by little,reduced food intake and water consumption,fur lost luster,reduced movement,loose stools,dirty tails etc.After intragastric administration,the low dosage,middle dosage and high dosage of Shengyang Yiwei Particles groups and the Nuodikang Capsule group rats' food and drinking intaking and movement were both increased,fur getting luster,feces getting normal.The middle dosage and high dosage of Shengyang Yiwei Particles groups increased significantly.The change of model group was not significantly.(2)Weight and Behavioural Indicators: before modeling,each group rats' weight and behavioural indicators had no significant differences(P>0.05).After modeling,compared with the normal group,the other 5 model groups had significant differences(P<0.05),and there were no significant differences between the other 5 model groups(P>0.05).After intragastric administration,the model group rats' weights and behavioural indicators were less than the other 5 groups,which had significant differences(P<0.05).The normal group rats' weights and behavioural indicators were higher than Nuodikang Capsule group and the low dosage of Shengyang Yiwei Particles group,which had significant differences(P<0.05),but compared with the middle dosage and high dosage of Shengyang Yiwei Particles group,which had no significant differences(P>0.05).The middle dosage of Shengyang Yiwei Particles group compared with the high dosage,which had no significant differences(P>0.05).(3)The contents of SDH of rat skeletal muscle mitochondria: The model group rats' contents of SDH of rat skeletal muscle mitochondria were less than the normal group,which had significant differences(P<0.05);The model group rats' were less than Nuodikang Capsule group and the low?middle and high dosage of Shengyang Yiwei Particles group,which had significant differences(P<0.05);The Nuodikang Capsule group compared with the low?middle and high dosage of Shengyang Yiwei Particles group,which had significant differences(P<0.05).The middle?high dosage of Shengyang Yiwei Particles group compared with the low dosage,which had significant differences(P<0.05).The middle dosage of Shengyang Yiwei Particles group compared with the high dosage,which had no significant differences(P<0.05).(4)The contents of Cyt-c of rat skeletal muscle mitochondria: The model group rats' contents of Cyt-c of rat skeletal muscle mitochondria were less than the normal group,which had significant differences(P<0.05);The model group rats' were less than Nuodikang Capsule group and the low?middle and high dosage of Shengyang Yiwei Particles group,which had significant differences(P<0.05);The low ? high dosage of Shengyang Yiwei Particles group compared with the Nuodikang Capsule group,which had significant differences(P<0.05);The middle dosage of Shengyang Yiwei Particles group compared with the Nuodikang Capsule group,which had no significant differences(P>0.05);The middle ? high dosage of Shengyang Yiwei Particles group compared with the low dosage of Shengyang Yiwei Particles group,which had significant differences(P<0.05);The high dosage of Shengyang Yiwei Particles group compared with the middle dosage of Shengyang Yiwei Particles group,which had significant differences(P<0.05).conclusion:(1)In this experiment,we take a composite method to construct chronic fatigue rats model.This composite method can be easily repeated and operated.Because this method has made rats with excessive movement and psychological stress fatigue,it can stimulate people in inquiring chronic fatigue syndrome.(2)Shengyang Yiwei Particles can get back the chronic fatigue rats' weight to a healthy,make the rats' swimming exhaustion time longer,increase the rats acrossing grid and standing movement times;Shengyang Yiwei Particles can significantly improve the fatigue syndrome of CFS rats.This result provides the experimental evidence for the treatment of CFS with Shengyang Yiwei Particles in clinical application.(3)Shengyang Yiwei Particles can significantly increase the content of SDH and Cyt-c in skeletal muscle mitochondria of chronic fatigue rats,regulate the body's mitochondria energy metabolism,and improve the body' ability to use oxygen,and play its anti-fatigue effect.It's maybe one of mechanisms of anti-fatigue benefits Shengyang Yiwei Particles.
