Objective:This design mainly focused on the basic pathogenesis of CFS.By prepareing the rat model with chronic fatigue,we observe Shengyang Yiwei particles on chronic fatigue rats general situation and behavioural indicators(exhaustion swimming time,across the grid and standing movement times),observe the effect of Shengyang Yiwei particles on the contents of PGC-1? and COX of chronic fatigue rat skeletal muscle mitochondria,in order to explore the anti-fatigue action mechanism of Shengyang Yiwei particles,and then provide experimental and theoretical evidence for the clinical application of Shengyang Yiwei particles to treatment CFS.Method:(1)Grouping,Modeling:60 SPF SD male rats,after adaptive feeding 3 days,we randomly divided into:normal group,model group,Nuodikang Capsule group,Shengyang Yiwei particles low dose group,Shengyang Yiwei particles middle dose group,Shengyang Yiwei particles high dose group,each group had 10 rats.The normal group without modeling and the other 5 groups carried modeling,time for 14 days.We adopted forced chronic bondage,exhaustion swimming and clip tail composite method to construct the model of rats with chronic fatigue.(2)Method of administration and dose: Ending the modeling,the normal group and the model group according to the 10ml/(kg · d)given normal saline,the Nuodikang Capsule group according to 0.182g/(kg · d)given Nuodikang Capsule suspension gavage,the Shengyang Yiwei particles low dose group according to 1.625g/(kg · d)given Shengyang Yiwei particles suspension gavage,the Shengyang Yiwei particles middle dose group according to 3.25g/(kg · d)given Shengyang Yiwei particles suspension gavage,the Shengyang Yiwei particles high dose group according to 6.5g/(kg · d)give Shengyang Yiwei particles suspension gavage.Rats in each group were 4 administered 1 times a day,for 14 consecutive days.(3)Extract tissue: After successful local anesthesia in rats,we took the piece of left hind limb skeletal muscle(1g)of rats in each group,and sinse in normal saline to prepare skeletal muscle tissue homogenate and mitochondria suspension,then packing label keeped in the-20? freezer.(4)The detecting of experimental indicators:We measured the rat body weight,exhaustion swimming time,across the grid and standing movement times in the day before modeling,after modeling and after the last administration.We evaluated the contents of PGC-1? and COX of rat skeletal muscle mitochondria by ELISA method.We used SPSS18.0 statistical software to process the collected data.Results:(1)General Situation: before modeling,there is no difference with eating and drinking,fur,movement and feces between each group of rats.After modeling,every modeled group rats reduced food intake and water consumption,fur lost luster,squinting lazy move,loose stools etc.The normal group rats eating and drinking as usual,fur bright color,normal activities and feces normal.After intragastric administration,the Nuodikang Capsule group,the low dosage,middle dosage and high dosage of Shengyang Yiwei particles groups rats' food and drinking intaking and activities were both increased,the fur became more glossy,the feces was back to normal.The middle dosage and high dosage of Shengyang Yiwei particles groups increased significantly.The change of model group was not significantly.(2)Weight and Behavioural Indicators: before modeling,each group rats' weight and behavioural indicators had no significant difference(P>0.05).After modeling,compared with the normal group,the 5 model groups had significant difference(P<0.01),and there had no significant difference between the 5 model groups.After intragastric administration,the model group rats' weights and behavioural indicators were less than the other 5 groups,which had significant difference(P<0.01).The normal group rats' weights and behavioural indicators were higher than Nuodikang Capsule group and the low dosage of Shengyang Yiwei particles group,which had significant difference(P<0.01),but compared with the middle dosage and high dosage of Shengyang Yiwei particles group,which had no significant difference(P>0.05).The middle dosage of Shengyang Yiwei particles group compared with the high dosage,which had no significant difference(P>0.05).(3)The contents of PGC-1? and COX of rat skeletal muscle mitochondria: The model group rats' contents of PGC-1? and COX of rat skeletal muscle mitochondria were less than the other 5 groups,which had significant difference(P<0.01).The normal group rats' were higher than Nuodikang Capsule group and the low dosage of Shengyang Yiwei particles group,which had significant difference(P<0.01),but compared with the middle dosage and high dosage of Shengyang Yiwei particles group,which had no significant difference(P>0.05).The middle dosage of Shengyang Yiwei particles group compared with the high dosage,which had no significant difference(P>0.05).Conclusion:(1)In this experiment,we take a composite method to caused both excessive movement and psychological stress(chronic bondage,exhaustion swimming and clip tail)to build the chronic fatigue rat model.This way is feasible and it can highly simulates the human body's chronic fatigue syndrome.(2)Shengyang Yiwei particles can get back the chronic fatigue rats to a healthy weight,significantly increase the rats acrossing grid and standing movement times,extend the swimming exhaustion time,and then improve their fatigue condition.This result provides a basic research data for the clinical application of Shengyang Yiwei particles treats CFS.(3)Shengyang Yiwei particles can significantly increase the content of PGC-1? and COX in skeletal muscle mitochondria of chronic fatigue rats.It's maybe one of mechanisms of anti-fatigue benefits Shengyang Yiwei particles.(4)The middle dosage of Shengyang Yiwei particles is the normal dosage of human body,increasing the dosage is not useful for it anti-fatigue effect.
