Objective:This topic based on the basic theory of TCM, Through by the experimental observation,to investigate the effects of Kidney phlegm prescription on PCOS rats of hormone levels andovarian tissue morphology,and revealed the mechanism of Kidney phlegm prescriptionhow to reduce the luteinizing hormone, androgen and the effect of improve themorphological changes in ovarian tissue in the PCOS rats. To provide a scientific basis forthe effectiveness of kidney phlegm PCOS Treatment.Methods:The topics chosen95the same species of clean grade21-day-old female SPFrats,the95SPF rats were randomly divided into70pathology model group and the other25rats which daily subcutaneous injection of large soybean oil as normal control group.Dailysubcutaneous injection of The70pathology model group rats dehydroepiandrosterone(DHEA)6mg/100g weight+0.2ml injection oil pathology group, continuous20d;TheControl rats injected daily subcutaneous injection of0.2ml of oil, continuous20d.After themodel, first estrous cycle determination and OGTT test, from the normal group and modelgroup rats were randomly selected10models were assessed; upon successful model in ratsto assess the next step experiment.There are60pathological model rats and15normal rats,the60pathological model rats were randomly divided into metformin group, low dose groupand high dosage groups of15normal as a group of15normal control group. After fourweeks of continuous gavage, according to the results of the vaginal smear, the firstperiod of fasting during the estrous but water12hours, weighed, chloral hydrate (10%)anesthesia, abdominal aortic blood serum was separated, and then sacrificed these ratsby cut cervical, ovarian tissue with4%paraformaldehyde, the line separating theserum hormone measurement, light microscopy changes in ovarian tissue morphology.Results:①the experimental animal general status change:rats in the model group by subcutaneous injection of dehydroepiandrosterone have several phenomenon: reducing ofactivity level, increasing of eating, drinking, and urine volume. But weight gain is notobvious. The above change with the increase of the gastric drug delivery days, normal grouprats’ weight gradually growth, while mental signs and activity response as usual. Afterintragastric administration, some symptoms have appeared among the part of traditionalChinese medicine group rats, for example, losing of weight, reducing of water intake andappetite.②The experimental results show that: the model rats of luteinizing hormone LHand androgen were higher than that in the normal group, metformin group, high-dose group,low dose group, and with more than two groups in terms of comparison, the difference wasstatistically significan(P<0.05); compared with the model group, high, low dose group andLH and testosterone decreased significantly(P<0.05); compared with metformin group,high, low dose group of poor results, with statistical significance(P<0.05); luteinizingbetween high, low dose treatment group formation known as the statistical significance(P>0.05). Each group of rats’ androgen level showed that there are no statisticalsignificance between high and low dose treatment group (P<0.05). Results indicate: thetraditional Chinese medicine group decreased testosterone, luteinizing hormone, and thetraditional Chinese medicine high dose group‘s effects even more better.③The Influence onthe morphology of testis in rats: normal group: It is obvious that the ovary follicles at allstages of development, such as primary oocytes, secondary oocyte, follicle cells growth. Theo ocytes with corona, which cell structure is of closely spaced. There are also some numberof granule cells and corpus luteum. Ovarian tissue in rats of model group: there aredilatations of follicle, cystic follicular oocytes or corona radiata disappeared. It is obviousthat there are some multiple vesicles which is actually empty.Because of granule cells less,the cell level decreased and presents loose arrangement. The normal forms of the variouscells were rare, not even a little bit. Multiple locking cells and oocyte corona radiatavanished are visible, while the number of corpus luteal cells is decrease. Observed inmetformin group rat ovarian tissue: A handful of vesicles cells were visible, all levelsfollicular cells are visible in the field of vision and in the number is increase, we saw theprimary oocyte, secondary oocyte, growing follicles and mature follicle,we also saw themature follicle theca were fully developed and the inner cell were more bigger. The coronaradiata and zona pellucida were visible in Oocytes. And we saw the number of granulosacells was increase, Shape closely arranged; the number of corpus luteum was also increase.Inthe high-dose prescription group rats ovarian tissue:We saw a handful of vesicle cells,including a small number of primary oocyte, secondary oocyte, follicle growth and mature follicles, the number of granule cells than the untreated group increased, and there is acertain number of corpus luteum. In the low-dose prescription group rats ovarian tissue: wesaw the number of vesicles cells decreased, the number of yellow body slightly increased. Inaddition, we also saw the growth of the follicle cells and a small minority atresiacells.Compare each experimental group microscopic follicular and luteal Number of levels:the number of follicles and corpus luteum levels of rats in the model group were lower thanthe normal group, the difference was statistically significant(P<0.05). Compared with themodel group rats, the number of follicles and corpus luteum both in the high and lowmetformin group is more bigger, the difference was statistically significant(P<0.05).Therats of Metformin group compared with the rats of both the high dose and the low dosetreated follicular and luteal count the number of levels the difference was statisticallysignifican(tP<0.05).Compared with the number of follicles and corpus luteum levels of thehigh dose group and the low dose group, the difference between them was not statisticallysignificant(P>0.05). Although not statistically significant, but there is increasing trend.Conclusions:①The Kidney phlegm prescription could improve the mechanism of reproductivefunction in rats.②The Kidney phlegm prescription could affecting the reproductivefunction of rat ovaries,thus stabilized the PCOS rat model of reproductive endocrineby directly act on the ovary, making the hormone secretion become normal.③Thisexperiment confirmed that the Kidney phlegm prescription could fundamentallyimprove ovarian function, and thus can significantly improve ovarian tissuemorphology, making tends to be normal ovarian function.
