OBJECTIVE: To study the effect of Shugan Bushen expectorant Chinese medicine on serum hormones in polycystic ovary syndrome rats and to observe the effect of Shugan Bushen expectorant Chinese medicine on serum TGF-?1 / smad transduction pathway and TIMPS / MMPS in ovariectomized rats with experimental polycystic ovary syndrome.To explore the effect of traditional Chinese medicine on improving polycystic ovary syndrome in rats and the effect and mechanism of ovarian fibrosis in rats Methods: Six healthy male SD rats were randomly divided into 6 groups: normal control group,model group,western medicine positive group,low dose group of traditional Chinese medicine treatment group,middle dose group and traditional Chinese medicine treatment group.In addition to the normal control group,the other five groups were treated with letrozole-carboxymethyl cellulose solution with concentration of 0.125 mg / ml to prepare polycystic ovary syndrome model,and at the same time The rats in each group were sacrificed and the abdominal aorta was sacrificed.The patients were sacrificed by enzyme-linked immunosorbent assay(ELISA)The levels of serum FSH,LH,E2,T and TIMPS and MMPS were observed.The pathological changes of ovaries were observed under light and electron microscope.The expression and distribution of TGF-?1 and smad3 were observed by immunohistochemistry.Western blot was used to detect the expression of TGF-?1,smad3 / 4 and smad7 in ovary of rats.Result: 1.Changes of serum hormone levels of FSH,LH,E2 and T in rats(1)the level of serum FSH in rats Serum FSH levels were highest in the blank group.Compared with the model group,the serum FSH serum levels of each group were significantly higher(P<0.01),Western medicine control group and low dose of traditional Chinese medicine group was significantly different(P<0.05),and Western medicine control group(P <0.05).There was no significant difference between the two groups(P> 0.05).There was no significant difference between the control group and the normal group(P<0.05)(P>0.05),but there was no significant difference between the two groups(P>0.05).(2)The expression of serum LH in ratsThe levels of serum LH in the control group were significantly lower than those in the control group(P<0.01).The level of serum LH in the model group was significantly lower than that in the control group(P<0.05),but there was no significant difference between the two groups(P> 0.05).However,there was no significant difference between the two groups(P>0.05).(3)Rat serum LH / FSH ratio comparisonCompared with the model group,the serum LH/FSH ratio was significantly higher in the model group than in the normal control group(P<0.01).However,there was no significant difference between the two groups(P> 0.05).However,compared with the normal control group,the middle dose of traditional Chinese medicine was significantly lower than that of the control group(P<0.01),but there was no significant difference(P> 0.05),so that in the improvement of LH/FSH ratio of Chinese medicine in the middle dose group the best results.(4)The expression of E2 in rat serumCompared with the normal control group,the level of serum E2 in the model group was significantly lower than that in the normal control group(P<0.01).Compared with the model group,the level of E2 in the serum of each group was significantly higher There was no significant difference in serum E2 level between the two groups(P>0.05).There was no significant difference between the two groups(P>0.05).There was no significant difference between the two groups(P>0.05).(5)rat serum T level expressionCompared with the control group,the serum T level of the model group was significantly higher than that of the control group(P<0.05),and the serum T level in the normal group was significantly higher than that in the control group There was significant difference between the two groups(P <0.05).2.TGF-?1 / smad transduction pathway:(1)The expression of TGF-?1 and its receptor was significantly higher in the model group than in the normal group(P<0.01).Compared with the model group,the expression of TGF-?1 and TGFThe expression of TGF-? 1 and TGF-? 1 in the dose group was significantly lower than that in the western medicine group(P>0.05).(2)Smad3 expression: Compared with the blank group,the expression of smad3 in the model group was significantly higher than that in the model group(P<0.01).Compared with the model group,the expression of smad3 in the low,middle and high group was lower(P<0.05).There was no significant difference in the expression of smad3 in low and medium group(P>0.05).(3)Smad4 expression: Compared with the blank group,the expression of smad4 in the model group was significantly higher than that in the model group(P <0.01).Compared with the model group,the expression of smad4 in the middle and high dose groups(P <0.05),and there was no significant difference between the two groups(P> 0.05).(4)Smad7 expression: Compared with the blank group,the expression of Smad7 protein in the model group was significantly lower than that in the blank group(F = 53.274,P<0.01).There was no significant difference between the middle dose group and the western medicine group(F = 43.432,P <0.01).Compared with the model group,there were significant differences in the RI values between the two groups(F = 43.432,P<0.01),and the RI of the middle dose group was statistically different(F = 36.854,P <0.05).The RI of the western medicine group was significantly different(F = 46.778,P <0.01).There was a significant difference between the two groups(P<0.01).3.TIMPS / MMPS system:(1)TIMPS expression: Compared with the normal control group,the expression of the model group increased significantly(P <0.01),compared with the model group,the expression of the treatment group decreased,the difference was statistically significant(P<0.01),And there was no significant difference between the low dose and the middle dose group(P>0.05).(2)The expression of MMPS: Compared with the normal control group,there was significant difference(P<0.01)in the model group(P<0.01).Compared with the model group,the expression of MMPS was significantly lower There was no significant difference in the level of serum MMPS between the two groups(P>0.05).There was no significant difference between the two groups(P>0.05).4.Ovarian ultrastructure Observation:(1)rat ovarian light microscope observation results: polycystic ovary ovarian ovarian cystic expansion of the follicle,was polycystic changes,showing primary follicles,atresia follicles and cystic expansion of follicles,granule cell layer decreased;After the treatment of kidney and expectoration,the follicles and mature follicles of different developmental stages were increased,and the number of granulocytes increased significantly,which indicated that the improvement of ovarian PCOS status and the recovery of ovulation function were observed after the treatment of Chinese medicine Shugan Bushen expectorant.(2)The results of electron microscopic observation of ovariectomized rats were observed by electron microscopy.The nuclei of the ovary granulosa cells were wrinkled and the nuclei were irregular.The nuclei of the ovary granules were agglomerated,chromatin was concentrated and collected.Vacuoles,mitochondria swelling,there are obvious morphological characteristics of apoptosis,Chinese medicine Shugan Bushen expectorant treatment of granulocytes can be seen after the normal nuclei and nucleoli,the nucleus chromatin uniform,multi-layer ovarian granulosa cells evenly distributed,cytoplasm Visible mitochondria rich,was long oval.in conclusion: 1.The model of PCOS rat induced by letrozole was successfully established.The model of ovarian morphology and endocrine hormone was studied.The results showed that the pathological morphology and serum hormone level were similar to those of PCOS.Therefore,An ideal animal model for the study of PCOS;2.Chinese herbal medicine Shugan Bushen expectoration method can improve the ovarian polycystic changes and ovulation disorder in PCOS rats;can improve the intracellular nuclei and mitochondrial status of ovarian granulosa cells in PCOS rats,reduce the number of glial cells in follicular cells and The number of lipid droplets,suggesting that it may improve the serum hormone levels in PCOS rats through the effects of the ultrastructure of granulosa cells and follicular cells;3.Chinese medicine Shugan Bushen expectoration by adjusting the endocrine level,reduce serum LH,T hormone levels,increased E2 hormone expression and reduce the LH / FSH ratio,to achieve the purpose of treatment of polycystic ovary syndrome;4.The expression of TGF-?1 and smad3/4 and the expression of smad7 were increased by inhibiting the expression of TGF-?1 / smads,and the expression of TGF-?1 and smad3/To achieve the purpose of anti-ovarian fibrosis.5.Chinese medicine Shugan Bushen expectoration method can reduce the expression of TIMPS in rat ovary,so that TIMPS / MMPS in a balanced state,to ensure the stability of ECM environment,to achieve the role of anti-ovarian fibrosis.
