Objective: To explore the effect of Qutan Huayu prescription on the model of Polycystic ovary syndrome(PCOS)in SD rats induced by dehydroepiandrosterone,to observe the challenge of Follicle stimulating hormone(FSH),Luteinizing hormone(LH),Testosterone(T),expression of ovarian TGF-?1 and thickness of granule cell layer,and to explore the mechanism of Qutan Huayu prescription in the treatment of polycystic ovary syndrome,so as to provide experimental basis for clinical application of Qutan Huayu prescription in the treatment of polycystic ovary syndrome.Methods: 80 female SD rats were randomly divided into blank control group(22 rats)and pathological model group(58 rats).Since the age of 22 days,the pathologic model group were given nape subcutaneous injection of dehydrogenation(DHEA)for 20 consecutive days,indicated that weight +0.2ml soybean oil mixture to establish PCOS model,the rats in the blank group subcutaneous injection of oil 0.2ml/d.After the modeling,10 rats in the model group and the control group were taken to carry out model evaluation.The rest of the blank group(12 rats)and model group(48 rats)carry on drug intervention.Rats in the pathologic model group were divided into high-dose group of Chinese medicine,low-dose group of Chinese medicine,Metformin group,and model control group.The blank control group,model control group,eat and drink freely.High dose group of Traditional Chinese medicine,low dose of Chinese medicine group and metformin group According to Bios' s method to calculate the dose of per kilogram weight,by body weight of the rats used to calculate the dose.Continuous administration for 28 days to observe each rat vaginal exfoliated cells smear;when in the same estrous cycle,fasting 12 h,anesthetized with 10% chloral hydrate 0.3ml / 100 g,collects the blood sample from abdominal aorta,separation of serum,preservation in-20 ?,ELISA reagent kit to detect the concentration of follicle stimulating hormone(FSH),luteinizing hormone(LH),testosterone(T)in serum.After taking the left ovaries of rats weighing and quickly placed in 4% of poly formaldehyde solution fixed.HE staining was used to observe the rat ovarian morphology,and measuring the thickness of follicular granule cell layer,by Masson staining method to observe the degree of ovarian fibrosis,the immunohistochemistry staining method detect the expression of TGF-?1.Results: 1.Model evaluation: estrous cycle:(1)vaginal smear of rats after 10 days shows: rats in the blank group cognizance vaginal opening day as the emotional period,24 hours after entering the metaoestrus,its regular estrous cycle is 4-5 days;rats in the model group,the changes of estrous cycle in irregular,extended to 8-9 days,or no obvious estrous cycle.(2)Significant difference of LH level between that in model group and blank group rat(P < 0.01);blank group compared with the model group,the FSH has differences(P < 0.05);serum T level in the model group compared with blank control group has difference(P < 0.05).(3)The level of serum INS in the rats of model group was significantly higher than that in normal group,P < 0.01,with significant difference.(4)Compared with the model group rats,the control group's left ovary show a different weight,P < 0.05,has statistical significance.The body mass index(BMI)of rats in the model group compared with the control group was not statistically significant,PD > 0.05.2 evaluation of after administration:(1)through the observation of vaginal smear of rats,the rats in the blank group with regular estrous cycle disorder;model group rat estrous cycle,irregular ovulation.Through drug intervention,high dose traditional Chinese medicine group,low dose traditional Chinese medicine group and metformin group rats' estrous cycle approximate recovery.(2)Compared with the rats in the model group,the other rats LH levels were significantly lower,with significant difference(P < 0.01).The level of LH in high dose group was higher than that in metformin group(P < 0.01),and the level of LH in low dose group was not significant(P < 0.05).Compared with the model group,the serum FSH level of rats in each group was significantly different,which was statistically significant(P < 0.05).Compared with model group,each rat serum T level were statistical significance(P < 0.05).(3)traditional Chinese medicine high dose,traditional Chinese medicine low dose,metformin group and model group rats' left ovary weight have statistical significance(P < 0.01),Chinese high,low dose group and metformin group had no difference,does not have statistical significance(P > 0.05).(4)Expression of ovarian TGF-?1,high dose group and blank group had no significant difference(P < 0.05)and the rest group compared with the control group were significant difference(P < 0.01);high dose of Chinese medicine group compared with the metformin group has statistical significance(P < 0.05),low dose of Chinese medicine group compared with dimethyl metformin group has no statistical significance(P > 0.05).(5)the high dose group's thickness of granule cell layer compared with the blank group has no statistical difference(P > 0.05),and the rest of the group compared with the blank group statistical significance(P < 0.01);high,low dose traditional Chinese medicine group compared with metformin group,showed no significant difference,does not have statistical significance(P > 0.05).Conclusion: 1.Qutan Huayu prescription can reduce PCOS rat LH levels and elevated FSH levels,improve PCOS rat model of endocrine,thereby restoring ovarian function,promote normal ovulation.2 Qutan Huayu prescription can decrease the expression of TGF-?1 in ovarian interstitial in PCOS rats,reduce the degree of fibrosis of ovary.3 Qutan Huayu prescription can lead the thickness of the follicular granular cell layer of the rat with polycystic ovary syndrome.
