Objective Through the statistics of hormone induced femoral head necrosis factorVEGFmRNA and BMP-2content of the treatment group and the control group in theanalysis, to explore the Liu Bai Chang Sha Decoction treatment of steroid inducedavascular necrosis of the femoral head mechanism, put forward theories, looking for atreatment of steroid induced avascular necrosis of the femoral head and effectivemethod for the clinical.Methods50experimental animal were randomly divided into blank control group(10rats), SANFH group (10rats), Xianlinggubao group (10rats), Liu Bai Chang ShaDecoction low dose group (10rats), Liu Bai Chang Sha Decoction high dose group (10rats). Consult the molding method of Li Zirong. Experimental animal accurate weighing,except the blank control group, each with horse serum10ml/kg, the rabbit ear veininjection,2week interval, at a dose of5ml/kg for two consecutive days injection ofhorse serum (1daily injections),2week intervals for3consecutive days, intraperitonealinjection of methylprednisolone40mg/kg (1daily injections). Give the blank controlgroup normal saline3days continuous injection of equivalent. In the modeling of thestart time, E group of experimental animal every day by0.5g/Kg gavage, group Dexperimental animal every day by0.25g/Kg gavage, group C experimental animal everyday by0.25g/Kg gavage, blank control group and SANFH control group single cagefeeding, no treatment. After6weeks, by immunohistochemistry in Liu Bai Chang ShaDecoction on the glucocorticoid induced avascular necrosis of the femoral head in vivoVEGFmRNA and BMP-2effect.Result The general observation: in group A, the rabbit body weight increased, haircolor and moist, the activity is sensitive; group B, the weight does not increase, lowspirit, fluffy fur dull, without gloss, subcutaneous fat thickness, reduced activity, whenthe activity after lower limbs become more clumsy, bouncing time reduced height,jumping distance, individual rabbit appear anorexia,2rabbits died; group C, the rabbitbody weight increased slightly, the shaggy but glossy, flexible activity than that in B group,1rabbits died; group D, the rabbit body weight increased, the hair shiny,activities when the bounce distance than in the C group,1rabbits died; group E, therabbit body weight increased, normal diet, hair color and moist, flexible. The femoralhead pathology observation: A group of rabbit femoral head, femoral neck withcomplete shape, the bone surface smooth, hard bone, hyaline cartilage, the whole iswhite; B group of rabbit femoral head still has a complete form, but the femoral neckbecame crisp, easily broken, but found that part of the femoral head joint surfacecoarseness and collapse of pathological phenomena, the cortical thinning, femoral headcartilage lose luster, dark color, the blue; C group of rabbit femoral head is still intact,part of the articular surface of the femoral head appear coarse, cortical thinning, femoralhead cartilage glossy dark; D group of rabbit femoral morphology the basic integrity,bone hard, the articular surface of the femoral head was normal, shiny; E group ofrabbit femoral head bone morphology, surface smooth, hard bone, cartilage andtransparent, the overall color is pale white. The results of immunohistochemistry:animal VEGF mRNA positive stained area percentage and the severity of SANFH isclosely related to, the B group compared with other groups, P values were less than0.05,compared with C group and D group, P>0.05, compared with E group and C group, Dgroup, P values were less than0.05. The experimental animal VEGF mRNA positiveexpression of the average gray value and the severity of SANFH is closely related to,the B group compared with other groups, P values were less than0.05, compared with Cgroup and D group, P>0.05, compared with E group and C group, D group, P valueswere less than0.05. The experimental animal BMP-2positive stained area percentageand the severity of SANFH is closely related to, the B group compared with othergroups, P values were less than0.05, compared with C group and D group, P>0.05,compared with E group and C group, D group, P values were less than0.05. Thepositive expression of BMP-2in experimental animal average gray value and theseverity of SANFH is closely related to, the B group compared with other groups, Pvalues were less than0.05, compared with C group and D group, P>0.05, comparedwith E group and C group, D group, P values were less than0.05.Conclusion1Liu Bai Chang Sha Decoction can improve the expression ofVEGFmRNA and BMP-2in rabbit;2Liu Bai Chang Sha Decoction has a preventiveand therapeutic effect on necrosis of the femoral head of rabbit.
