| Objective:The topic takes activating blood circulation and tonifying the kidney to strengthen bone as the theoretical guidance, to observe the effects of Tongluo Zhuanggu Pill on the hemorheology indexes in rabbits of hormonal avascular necrosis of femoral head.Probing into its working mechanisms of preventing and curing of hormonal avascular necrosis of femoral head,providing theoretical basis for clinical treatment of hormonal avascular necrosis of femoral head of Tongluo Zhuanggu pill.Methods:The experiment used 26 healthy New Zealand rabbits as the experimental object. to choose 9 rabbits randomly as blank group,the others were estabed animal model. The horse serum 10 m L / kg injected through ear vein to the animal model group.After 3 weeks,the horse serum reduction changed to 6m L/kg ear vein injection again.After 2 weeks, according to 40mg/kg of methylprednisolone intraperitoneal injection to rabbits. 1 times a day, for 3 days.4 weeks after modeling,the blank group,modeling group randomly selected 1 rabbits were sacrificed after molding.Dissecting rabbit femoral head rapidly and abserving their appearance, shape, color and histopathological observation to confirm the modeling results.After confirming successful model,building modules were randomly divided into model group, treatment group, each group of eight.Control group, model group were fed once a day with saline 10 ml and treatment group was given the solution of 0.3g/kg/d Tongluo Zhuanggu pill 10 ml.All rabbits were treated with a single cage feeding by optional water,and test weight to adjust the dosage regularly.After 8 weeks of continuous treatment,the blood was collected from the abdominal aorta to measur the data of blood rheology.Then all the animals were sacrificed in experimental animal,and the blood rheology data using SPSS18.0 statistical analysis.Results:①Rabbit hair, modeling group activity, reaction, mental status, appetite and femoral head appearance were not better than the blank group,and the body weight was significantly lower than that of the control group,too.Early manifestations of treatment group was similar to model group rabbits,but after 2 weeks of treatment, the performance began to improve,and hair, activity, reaction, mental status, appetite and femoral head were close to the normal group in the sampling time,and the weight is also similar before the experiment.②Under light microscope:No abnormal change in the normal group,but the trabecular bone was thinning, breaking obviously of the model group,and the marrow fat cell is hyperplasia and hypertrophy.The treatment group pathological change is not obvious and the trabecular bone structure is basically complete and orderly arrangement.The proportion of medullary cavity of hematopoietic cells and fat cells is close to the normal.③The results of blood rheology:Indexes of blood rheology generally increased in model group.The whole blood viscosity(high shear, low shear),plasma viscosity,hematocrit,whole blood reduction viscosity(high shear, low shea) of the model group were increased significantly(P < 0.01).The index of treatment group blood rheology indicators is generally reduce.There is significant difference in every index of blood rheology(P < 0.01) and close to the blank group(P>0.05) compared with the model group.Conclusion:Tongluo Zhuanggu pill can reduce the whole blood viscosity,plasma viscosity,hematocrit, the whole blood reduction viscosity(high shear, low shear) of the steroid-induced femoral head osteonecrosis of rabbit.By improving the permeability of capillaries to improve the femoral head microcirculation,ensure the effective perfusion of femoral head,and promote the dead bone absorption, accelerate the necrotic bone repairing, promote new bone regeneration effect.Tongluo Zhuanggu pill is significant for steroid-induced femoral head osteonecrosis prevention and treatment effect. |
