Objective:Through animal experiment,to analyse the pathogenesis of glucocorticoid—induced femoral head necrosis,and after hormone application given medicine Hong-HD particles intervention,by observing the indicators of triglyceride (TG), total cholesterol (TC), high-density lipoprotein cholesterol (HDL), low-density lipoprotein cholesterol (LDL), high shear blood viscosity, low shear blood viscosity,erythrocyte sedimentation rate (ESR), Plasma viscosity in serum,and the H-E staining biopsy of the femoral head,for the the Hong-LD particles prevention femoral head necrosis to provide experimental basis.Methods:There are healthy adult rabbits45, weight2.4+0.5Kg.They are randomly divided into three groups,15in each group.Group A:given standard diet and saline5ml orally every other day for2weeks at room temperature, for experimental control; Group B:Given5ml prednisone acetate solution (including prednisone acetate4.6mg/kg) orally every other day for2weeks;Group C:Given5ml acetate solution (including prednisone acetate4.6mg/kg and Hong-LD particles2g) orally every other day for2weeks. To exsanguinate interval from the marginal ear vein used for lipids and hemorheology laboratory detection after the experimental animals into the animal room week later,and exsanguinate interval marginal ear vein for laboratory testing again four weeks after the drug treatment.The experimental animals were sacrificed by taking air embolism, removed the femoral head.Three groups femoral head trabecular bone, bone cells and its changes were observed by light microscopy after H-E staining.The resulting data using IBM SPSS Statistics19statistical package handling,were expressed as meanJ: standard deviation, to take a completely randomized design analysis of variance for statistical analysis.Results:1.The indicators of lipids:(1) Within-group comparisons:No significant difference between group A before and after treatment.After treatment,TG, TC, LDL was significantly higher, and HLD significantly reduced in Group B,the difference was statistically significant (P<0.05).In Group C,LDL has no significant change after treatment, TG, TC was increased, HLD was decreased, but the difference was not statistically significant (P>0.05).(2)Comparison between groups:The TG, TC, LDL, HDL were not statistically different of three groups before treatment.After treatment,the difference was statistically significance (P<0.05) between Group A and group B, the difference of TG、TC、LDL was statistically significance (P<0.05) between Group B and group C, HDL in Group C was significantly higher than that in group B, but not statistically significant (P>0.05).2.The indicators of hemorrheology:(1) Within-group comparisons:The G (200/s), G (3/s), ηρ, ESR were increased in3groups after treatment,the difference was not statistically significant (P>0.05) in Group A and group C,but the difference was statistically significance (P<0.05) in Group B.(2)Comparison between groups:The G (200/s)、G(3/s)、ηρ、ESR were not statistically different in three groups before treatment.Group B was significantly higher than that in group A and group C,after treatment,and the difference was statistically significant (P<0.05)in group B and group A, group B and group C. The difference between Group C and group A was not statistically significant (P>0.05).3.Pathological observation:(1) Light microscope:In Group A,trabecular bone structure is neatly arranged rules, the number of bone cells in the most, mostly round nucleus in the center, around the trabecular bone more osteoblasts;In Group B,trabecular arranged scattered, bone cells is at least in three groups, trabecular bone atrophy, rupture, trabecular bone volume increases, karyopyknosis, partial located on one side of empty lacunae increased significantly;And in Group C,trabecular bone structure are somewhat sparse, the number of trabecular bone cells significantly more than group B, nuclei were larger, multi-partial located on the side of the visible part of empty lacunae.(2)The trabecular cell number and the percentage of empty lacunae:Trabecular cell count:cells in vision of Group A is more than group B and group C, and the difference was statistically significant (P<0.05) in group A and group B,but the different is not statistically significant (P>0.05)in Group A and Group C.Cells of group C is more than in group B,but the difference is not statistically significant (P>0.05).The empty lacunae percentage:The empty lacunae percentage of Group B was significantly higher more than that in Group A and Group C, and the difference was statistically significant (P<0.05), and it in Group C was more than that in group A, but the difference was not statistically significant (P>0.05).Conclusion:1, In the short to medium term given rabbits oral hormone can cause femoral head necrosis;2, The pathogenesis of steroid-induced osteonecrosis of the femoral head may have associated with serum fat metabolism, blood rheology;3,The Hong-LD particles can effectively delay or inhibit the development of the animal osteonecrosis of the femoral head occurred, and prevent animal femoral head necrosis.
