Purpose: The experimental study was designed to observe the bushencupaixilieprescription on androgen sterility rat,morphological changes of ovarian tissue andTNF-ɑ,EGF,TGF-1,IL-1,and to investigate the effective mechanism ofbushencupaixulie prescription further, so as to provide wider view of research andclinical guidance in the treatment of anovulatory infertility.Methods: we adopt healthy female SD rats of9-day age from the same batch inour research.Androgen sterility rat model was established by injectingtestosterone propionate subcutaneously. Experimental animals were divided intoseven groups randomly: high-dose group of bushencupailuan, low-dose group ofbushencupailuan,high-dose group of bushentianjing, low-dose group ofbushentianjing, clomiphene capsule group, blank control group and model controlgroup. Under the same conditions, they were fed in allotted cage. Gavaged Dailyonce. Bushencupailuanxilie prescription groups were respectively givenbushentianjing prescription29g/kg (for crude drug content),14.5g/kg,bushenhuoxue prescription24g/kg,12g/kg. Each prescription shall continuously begiven for3days in turn, six days is a cycle. Bushentianjing prescription groupswere respectively given29g/kg,14.5g/kg, respectively, equivalent to6.7times theclinical dose,3.35times. Clomiphene capsule group was given clomiphene capsule5.58mg/kg, which were made according to6.7times the clinical dose. The volumeof medication in each group was20ml/kg, the blank control group and the modelcontrol were given normal saline. The medicine was administered for30days.30days later, rats in the estrus were selected by vaginal smears, ELISA method wasused to measure the levels of TNF-ɑ,EGF,TGF-1,IL-1and observation of ovariantissue rats morphological changes by immunohistochemical method. Results:(1)the primordial follicles of high-dose group of bushencupailuan andclomiphene capsule group were increased significantly compared with the modelcontrol group(P<0.05). the number of primary and secondary follicles ofhigh-dose group of bushencupailuan, high-dose group of bushentianjing andclomiphene capsule group was increased significantly compared with the modelcontrol group(P<0.05). the number of mature follicles and corpus luteum ofhigh-dose group of bushencupailuan, high-dose group of bushentianjing andclomiphene capsule group was increased significantly compared with the modelcontrol group(P<0.01orP<0.05).(2)the significant differences of TNF-ɑ, EGF,TGF-1, IL-1were shown between the control model group and the bank controlgroup(P<0.01). the TNF-α of high-dose group of bushencupailuan, high-dose groupof bushentianjing and clomiphene capsule group was decreased significantlycompared with the model control group(P<0.01orP<0.05). the TNF-αof high-dosegroup of bushencupailuan were improved significantly(P<0.01),compared with thethe group of low dose of bushentianjing,and the differences between the high-dosegroup of bushencupailuan and the group of high dose of bushentianjing(P<0.05).thedifferences between high-dose group of bushencupailuan and clomiphene capsulegroup were not ststistically significant. the expression of EGF of the high-dosegroup of bushencupailuan, high-dose group of bushentianjing and clomiphenecapsule group decreased markedly(P<0.05),compared with the model control group.the expression of EGF of the high-dose group of bushencupailuan was obviouslylower than that of low-dose group of bushentianjing.and compared with thehigh-dose group of bushentianjing,there was no statistics difference, but thetendency reduced. The siginificant difference was shown in the both group ofbushencupailuan(P<0.05). the expression of TGF-β1of the high-dose group ofbushencupailuan and clomiphene capsule group decreased markedly(P<0.01),compared with the model control group. there were Statistical differencesbetween high-dose group of bushentianjing and the model control group. Comparedwith model control group, high-dose group of bushencupailuan, clomiphene capsulegroup、high-dose group of bushentianjing tended to decrease siginificantly in IL-1βexpression (P<0.01or P<0.05).the IL-1βexpression of high-dose group ofbushencupailuan, high-dose group of bushentianjing and low-dose group ofbushencupailuan the difference was statistically significant(P<0.05).Conclusion: Bushenpupailuanxilie can improve the morphological structure of the ovaries in pathological state, promote the follicles development and ovulation.The high pression of Ovary cell factors such as TNF-ɑ, EGF, TGF-1, IL-1participate in the pathogenesis of anovulatory disease.Bushenpupailuanxilie can regulate the higher levels of androgen sterility rats, so asto promote the follicles development and ovulation.
