| Objective:Based on previous research, observe the Tongmai Dasheng pill influence on renal deficiency induced anovulation infertility disorder rats. Through the role of BuShenTiaoChong,proposed Tongmai Dasheng pill mechanism of renal deficiency induced anovulation infertility and provide a better theory to guide clinical.Methods:Intramuscular injection of testosterone propionate used to model of renal deficiency induced anovulation infertility disorder rats. Through the observation of rat vaginal exfoliated cells. Selected successful model the rats and randomly divided into seven groups, control group, model group, the Tongmai Dasheng pill high-dose group, Tongmai Dasheng pill medium-dose group, Tongmai Dasheng pill low-dose group, Clomiphene citrate group,Yougui pill group. The rats were given the appropriate drug intervention. Observed after treatment of different groups generally clear conditions, the uterine index, ovarian index and its morphology, determination of the content of follicle stimulating hormonk and its receptor(FSH,FSHR),luteinizint hormone and its receptor(LH,LHR) and transforming growth factor-β1(TGF-β1),matri metalloproteinases 9(MMP-9).Results:1,By observing,normal rats have hair shiny:Model rats appear vertical hair, dull or yellow fur and often have alopecia; The drug intervention group compared with the model group to reduce the phenomenon of verticalhair and hair removal,but the difference was not significant.2,Normal uterus were thick volume,color red, endometrial thickness, the more number of glands, rich in blood vessels in the connective tissue.ovary were red and had more mature follicles and luteinizing, a high index uterine and ovarian index: Model uterus were small volume,pale color, endometrial thin,less glands.no significant secretion phenomenon.ovarian surface saw more pathological cystic follicles and no mature follicle and corpus luteum.a low index uterine and ovarian index. All drug intervention group no matter the index uterine and ovarian index or morphology showed significant recovery trend and had the mature follicle and corpus luteum formation under the microscope.3,The contents of FSH in rat serum and FSHR in ovarian tissue in model group were lower than that in nomal group,There were significant differences(FSH<0.05,FSHR: P<0.01). The contents of the intervention group increased and the Tongmai Dasheng pill high-dose group were higher than the other two groups, There were significant differences(Tongmai Dasheng pill medium-dose group:P<0.05, Tongmai Dasheng pill low-dose group:P<0.01).4,The contents of LH and LHR in model group were lower than the normal group(P<0.05) and another drug intervention group. In the Tongmai Dasheng pill All dose groups,the contents of LH in Tongmai Dasheng pill low-dose group were higher than that of Tongmai Dasheng pill high-dose group and Tongmai Dasheng pill medium-dose group. There were significant differences(P<0.05). The contents of LHR in Tongmai Dasheng.,pill high-dose group than the other two groups increased.but there were not significant differences(P>0.05).5,Compared with model group,there were control group,the Tongmai Dasheng pill high-dose group,Clomiphene citrate group,Yougui pill group.They had high level TGF-β1.The difference was significant(P<0.01),Tongmai Dasheng pill medium-dose group and Clomiphene citrate group can increase the level of MMP-9 too(P<0.05). In the Tongmai Dasheng pill All dose groups,the effect of the Tongmai Dasheng pill high-dose group and Tongmai Dasheng pill medium-dose group higher TGF-(31 than Tongmai Dasheng pill low-dose group.6,The contents of MMP-9 in model group were Significantly lower than that in model group. There were significant differences(P<0.01). After intervention by medication, The contents of Tongmai Dasheng pill high-dose group and Yougui pill group were increased significantly, There were significant differences(P<0.01). Tongmai Dasheng pill medium-dose group. The contents of Tongmai Dasheng pill low-dose groupand Clomiphene citrate group were slight increased, There were significant differences(P<0.05).Conclusion: 1,Renal deficiency induced anovulation infertility disorder rats were successfully established intramuscular injection of testosterone propionate.The model can lead to poor development of ovary and uterus, FSH, LH and their receptors are expressed low, Ovarian tissue expression of TGF-β1 and MMP-9 also significantly decreased.2,Through the pre-test and the results show that Tongmai Dasheng pill not only improve the uterus, ovarian morphology to some extent, but also reduce the pathologic cystic follicle number,increase amounts mature follicle and corpus luteum formation, promote the growth of ovums, development and discharge.3,Tongmai Dasheng pill can increase the level of pituitary gonadotropin and the level of the corresponding receptor in ovarian tissue.So,it not only improve the endocrine function of pituitary,but also improve the ovarian response to hormone stimulation. It makes the pituitary-ovarian endocrine function was higher exertion. For the local factors for ovarian, Tongmai Dasheng pill can improve the local micro-environment of ovarian by increasing low TGF-β1 and MMP-9 levels in renal deficiency induced anovulation infertility disorder rats.4,By endocrine regulation of gonadotropin and ovarian regulation of local factors, Tongmai Dasheng pill not only promote the growth of ovums, development,but also play an important role in mature follicle rupture, ovums excretion.
