Objective:5-Fluorouracil is a chemotherapy medicine witch has been widely usedin clinic application. This medicine can restrain the growth of cancer cells byinterrupting nucleic acid synthesis. Because of it has no specificity,5-Fluorouracilalways induces many complications such as intestinal mucosa injury and immunityrestrain. Probiotics can adjust intestinal flora and protect mucosa. In this decade,probiotics have been applied in clinic therapy. In this study, we have studied theprotective effect to5-fu induced intestinal mucosa injury and the security of acompound probiotics which contains Bifidobacterium breve, Lactobacillus acidophilus,Lactobacillus casei and Streptococcus thermophilus.Method:(1) The protective effect research:28SD rats were randomly divided intonormal group,5-Fu+saline group,5-Fu+high dose and low dose probiotics groups,7rats in each group. All rats were killed3days after chemotherapy. The change ofintestinal flora was detected by PCR-DGGE, plasma endotoxin was measured bytachypleus amebocyte lysate, the change of intestinal mucosa and splenic tissue weredetermined by HE staining.(2) The detecting of hazardous substance to probiotics:nitrate reductase was detected by NR testing kit; amino acid decarboxylase was detectedby BASM medium and hemolysin was detected by blood plant.(3) Drug resistanceassessment tests: the antibiotic susceptibility test performed to detect the level ofantibiotic sensitivity and agarose gel electrophoresis test to identify whether thetransferable resistant plasmid exist.(4) Acute toxicity test:20SD rats were divided intotwo groups, ten rats in each group. The experimental group was administrated with a large dose of experimental strains liquid culture; the control group was administratedwith equal normal saline. Then we observed the change of their habitus and mental state.We anatomized the mice at the end of observation to detect whether the structuraldisease happened.Result:(1) In protective effect study: Compared with5-Fu+saline group, theintestinal flora disturbance and weight lose have been improved (P<0.05), the level ofplasma endotoxin and intestinal mucosa injury have been significantly reduced (P<0.01)and the damage effect of5-Fu on spleen have been lightened.(2) Streptococcusthermophilus and Lactobacillus acidophilus can product amino acid decarboxylase, theother probiotics can not. All of probiotics in this study can not product nitrate reductase,hemolysin or plasmid.(3) In acute toxicity test, rats which have been administrated withprobiotics have gained more weight than control group (P<0.05). All the groups have nodeaths. Diarrhea, bacterial translocation and structural disease did not been observed.The change of MDA and GSH concentration has no differences between control andexperimental group (P>0.05).Conclusion: This compound probiotics showed good safety and can be effective inimproving rat intestinal mucosa, intestinal flora, immunity and growth condition after5-Fu chemotherapy.
